摘 要:本试验旨在探究不同剂量外源性PGF对小鼠黄体(corpus luteum, CL)溶解效果的影响及其剂量依赖性关系。选用60只60日龄、体质量30~34 g雌性昆明小鼠随机分为6组,分别采用腹腔注射1,10,50,100,200μg PGF。通过H.E染色法监测卵巢的形态学变化,ELISA法检测小鼠血清孕酮水平及利用qRT-PCR检测孕酮合成关键酶/细胞凋亡相关基因表达情况。结果显示,与对照组相比,不同剂量PGF处理组中昆明小鼠的CL溶解效果为50μg组>10μg组>100μg组>1μg组>200μg组;CL功能性指征结果显示PGF在50μg剂量范围内,小鼠体内P含量会随着PGF处理剂量的增加而呈现递增趋势;而且,P合成关键酶基因检测结果显示StAR和3β-HSD表达量在PGF注射剂量为50μg时呈现最低水平;同时,CL结构性指征检测结果发现此时促凋亡因子BAX基因、BAX/BCL-2比率及Caspase-3基因表达呈极显著性上调(P<0.01),而抗凋亡BCL-2基因呈极显著性下调(P<0.01)。结果表明,外源性PGF在调控CL溶解时50μg存在一定的临界值范围,即在该临界值范围内,PGF促CL溶解效果与其剂量间呈现明显的依赖性关系,这为后续探析雌性动物繁殖技能障碍(如持久CL)及优化PGs主导的高效繁殖技术方案研究奠定坚实的理论基础。The purpose of this study was to investigate the effect of different doses of exogenous PGFon the luteolytic effect of mice and the dose-dependent relationship between them.Sixty 60-day-old female Kunming mice weighing 30-34 g were randomly divided into 6 groups, and were injected intraperitoneally with 1 μg, 10 μg, 50 μg, 100 μg and 200 μg PGF,respectively.The morphological changes of the ovaries were monitored by H.E staining, the serum progesterone level in mice were detected by ELISA,and the expression of key enzymes in progesterone synthesis/apoptosis-related genes were detected by qRT-PCR.Compared with the control group, the results showed that the luteolytic effect in different PGFtreatment groups was 50 μg group>10 μg group>100 μg group>1 μg group>200 μg group.Results of luteal function indications showed that the Plevels in mice were increased with the increase of the dose of PGFwhen it is less than 50 μg.Moreover, the results of Psynthesis key enzyme genes showed that the expression levels of StAR and 3β-HSD were the lowest levels when the injection dose of PGFwas 50 μg.At the same time, the corpus luteum structural indication results showed that the pro-apoptotic factor BAX gene, BAX/BCL-2 ratio and Caspase-3 gene expression were significantly up-regulated at this time(P<0.01),while the anti-apoptotic BCL-2 genes were significantly down-regulated(P<0.01).These results show that there is a certain threshold range when exogenous PGFregulates luteolysis at 50μg.Within this threshold range,the luteolytic effect of PGFhas a significant dose-dependent relationship between them.This would lay a solid theoretical foundation for the subsequent analysis of reproductive skills barriers(such as persistent corpus luteum)in female animals and the optimization of PGs-led high-efficiency reproductive technology solutions.
