摘 要:本研究针对吉林省多地发生的一种新生犊牛先天性畸形和脑积水,临床上以四肢关节弯曲呈蜷缩状、无法伸直、不久发生死亡为特征的疾病进行了流行病学调查及病原分离与鉴定。结果应用MDBK细胞从发病与病死动物体内分离出3株病毒,命名为JL-AKA-CC21-1株、JL-AKA-CC21-2株和JL-AKA-YJ21。电镜负染观察显示,分离病毒粒子大小为70~130 nm,与病料中观察的病毒粒子相同。病毒接种MDBK细胞可引起典型的细胞病变,病变细胞聚集、变圆与脱落。分子生物学鉴定结果表明,上述分离毒株均为阿卡班病毒(Akabane virus, AKAV)。本研究从发生新生犊牛先天性畸形和脑积水的病死牛体内分离出AKAV,该研究结果将对今后吉林省本病的防控提供理论依据。In this study, we performed an investigation on the outbreak of unknown disease of newborn calves characterized by congenital abnormalities of arthrogryposis and hydranencephaly, and isolated three field isolates named JL-AKA-CC21-1,JL-AKA-CC21-2 and JL-AKA-YJ21 from sick or dead calves in different region of Jilin Province using MDBK cells.Electron microscopy observations showed the isolated virus particles were ranging from 70 nm to 130 nm in size, which were similar to the virus particles observed in the tissue specimens.The three virus isolates JL-AKA-CC21-1,JL-AKA-CC21-2 and JL-AKA-YJ21 caused a similar typical cytopathic effect in MDBK cells characterized by cell aggregate, become rounded and detached from flask.Molecular characterization showed all three virus isolates belong to Akabane virus.Taken together, the above results revealed that the Akabane virus was likely the main viruses responsible for the emerging infection in neonatal calves characterized by congenital malformalities of arthrogryposis and central nervous systems,thus providing the basis for the prevention and control of the disease in the future.
