摘  要:为分析2018—2020年豫南地区猪流行性腹泻病毒(porcine epidemic diarrhea virus, PEDV)的分子遗传变异情况,本研究利用RT-PCR方法从18个猪场PEDV感染仔猪组织中扩增S1基因部分片段、ORF3基因和N基因,进行核苷酸序列测定,并将PEDV流行株S1基因部分序列、ORF3基因和N基因推导氨基酸序列与参考毒株进行进化树构建与遗传变异分析。测序结果表明,18个猪场存在5株PEDV流行毒株。S1基因部分片段与N基因推导氨基酸进化树与相似性结果表明,5株PEDV流行株与已发表的2010年以后流行株组成G2群,与CV777株为代表的G1群毒株相似性分别为93.0%~96.7%和95.0%~96.8%。ORF3基因推导氨基酸进化树与相似性结果表明,5株PEDV流行株同其他非基因缺失毒株组成G2群,并分别属于G2a(HNXC和HNHC株)和G2c亚群(HNHB、HNNY和HNZY株),与缺失毒株组成G1群毒株的相似性为88.0%~92.4%,与CV777毒株所在G2b亚群毒株相似性分别为94.7%~96.4%和94.7%~96.9%。氨基酸遗传变异分析结果表明,与经典毒株CV777相比较,5株PEDV流行株的S1部分氨基酸、ORF3氨基酸和N氨基酸分别存在10,5,10个位置发生相同氨基酸置换,以及分别有16,10,14个位置氨基酸不同程度的突变,此外,5株PEDV流行株ORF3氨基酸不存在缺失,具有PEDV强毒株的分子遗传特征。综上所述,豫南地区PEDV流行具有PEDV变异毒株及强毒株的分子遗传特征,可为该地区PED的防控提供理论指导。To investigate the molecular genetic variation of porcine epidemic diarrhea virus(PEDV) in southern Henan from 2018 to 2020,the partial S1 gene, ORF3 gene and N gene were amplified from tissues of PEDV-infected piglets of 18 pig farms by RT-PCR,and sequenced.Phylogenetic relationships and genetic variations were analyzed with other PEDV reference strains based on the deduced amino acid sequence of partial S1,ORF3 and N genes.Sequence analyses revealed 5 PEDV epidemic strains from 18 farms in southern Henan.Phylogenetic and similarity analysis of partial S1 and N genes showed that the 5 PEDV epidemic strains, together with the published strains after 2010,were classified into the G2 group.They had similarities with the G1 group(including CV777 strain and other published strains before 2010)of 93.0%-96.7% and 95.0%-96.8%,respectively.Phylogenetic tree and similarity analysis of ORF3amino acid indicated that 5PEDV epidemic strains were classified into G2group with other non-gene deletion strains(HNXC and HNHC strains in G2,HNHB,HNNY and HNZY strains in G2csubgroups),and the similarity with G1group strain(including the deletion strain)was 88.0%-92.4%,and the similarity with G2bsubgroup(including CV777strain)were 94.7%-96.4% and 94.7%-96.9%,respectively.Compared with the amino acid sequence of CV777strain,there were amino acid substitutions at position 10,5and 10and mutations at position 16,10 and 14 for S1partial protein,ORF3protein,and N protein of the 5PEDV strains,respectively.In addition,no amino acid deletion was found in ORF3 of 5PEDV epidemic strains,which showed the molecular genetic characteristics of PEDV virulent strains.In summary,the PEDV epidemic strains showed the molecular genetic characteristics of PEDV variant strains and virulent strains,which can provide theoretical guidance for the formulation prevention and control measures of PED in southern Henan Province.
