摘 要:目的基于网络药理学和分子对接技术,预测体外抗菌效果较好的中药活性分子治疗肺结核的潜在作用机制。方法通过文献筛选出体外抗结核分枝杆菌效果较好的中药活性成分,利用分子对接技术、KEGG通路、GO功能分析等方法进行探究活性成分治疗肺结核潜在机制。结果姜黄素、黄芩苷、虫草素、穿心莲内酯、白黎醇素、木犀草素较筛选的其他中药活性成分在治疗肺结核病方面效果更好。主要通过肺结核通路、IL-17信号通路、吞噬体通路和NF-κB转录因子活性的正向调节、JAK-STAT级联的正调节、对细菌源性分子的反应等功能发挥治疗肺结核的作用。结论12种中草药活性成分可能通过多条通路、多种代谢途径产生抑制炎症反应、免疫功能调节、提高机体免疫力、提高对结核分枝杆菌杀伤力等作用来达到治疗肺结核病的目的。Objective To predict the potential mechanism of traditional Chinese medicine active molecules with good antibacterial effect in vitro in the treatment of pulmonary tuberculosis based on network pharmacology and molecular docking technology.Methods The active components of traditional Chinese medicine with good anti mycobacterium tuberculosis effect in vitro were selected through the literature,and explored the potential mechanism of active ingredients in the treatment of pulmonary tuberculosis by molecular docking technology,KEGG pathway and GO function.Results Curcumin,baicalin,cordycepin,andrographolide,bailitol and luteolin were found to be more effective in the treatment of pulmonary tuberculosis than other active ingredients of traditional Chinese medicine.It is mainly through pulmonary tuberculosis pathway,IL-17 signaling pathway,phagosome pathway and positive regulation of NF-κB transcription factor activity,the positive regulation of JAK-STAT cascade,and the response to bacterial derived molecules that they play a role in the treatment of pulmonary tuberculosis.Conclusion The active ingredients of 12 kinds of Chinese herbal medicines may inhibit inflammatory reaction,regulate immune function,improve body immunity,and increase the killing power to mycobacterium tuberculosis through multiple pathways and metabolic pathways to achieve the purpose of treating tuberculosis.
