摘 要:目的探讨黄葵胶囊治疗糖尿病肾病的作用机制。方法通过TCMSP、GeneCards、Swiss Target Prediction以及文献检索等手段筛选出黄葵胶囊的活性成分、作用靶点及糖尿病肾病作用靶点;采用String数据库对药物与疾病共有靶点构建蛋白互作网络图(PPI),利用Metascape数据库对共有靶点进行GO、KEGG富集分析,Cytoscape(3.7.1)软件构建“药物-成分-靶点-通路”网络。采用动物实验验证黄葵胶囊对核心靶点的调节作用。结果筛选出47个关键靶点,包括HIF1A、GAPDH、VEGFA、TNF、STAT3等。动物实验验证了黄葵胶囊可能通过降低TNF-α、VEGFA等核心靶点的表达量,调节炎症反应,从而降低糖尿病肾病小鼠的尿蛋白。结论黄葵胶囊可能通过下调TNF-α、VEGFA等蛋白的表达,在糖尿病肾病的治疗中发挥作用。Objective To explore the mechanism of Huangkui capsule in treating diabetic nephropathy.Methods The active components of Huangkui capsule were searched through traditional Chinese medicine database and literature review,and their targets were predicted by Swiss target prediction.The targets of diabetic nephropathy were obtained through GeneCards database.The common targets were selected to construct Protein Protein Interaction network(PPI)through string database,and screened by Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)according to metascape database.The network of“drug-component-target-pathway”was constructed through Cytoscape(3.7.1)software.Animal experiments were used to verify the regulatory effect of Huangkui capsule on core targets.Results There were 47 key targets,including HIF1A,GAPDH,VEGFA,TNF,STAT3,etc.Animal experiments had verified that Huangkui capsule could modulate the inflammatory reaction through the expression of TNF-α,VEGFA and other core targets,and reduced the urine protein of diabetic nephropathy mice.Conclusion Huangkui capsule mainly down the expression of TNF-α,VEGFA and other targets,play a role in the treatment of diabetic nephropathy.
