摘 要:研究西藏林芝市察隅县水稻根际土壤丝状真菌群落结构,探究影响其多样性的理化因子.通过稀释涂布平板法分离丝状真菌,结合经典分类法、rRNA-ITS转录间隔区序列分析对丝状真菌进行系统分类,测定土壤理化指标,综合分析丝状真菌多样性与理化因子的相关性.最终从察隅县水稻根际土壤中分离纯化出221株丝状真菌,归为17个属33个种,其中优势属为Penicillium,包括10个种,且与铵态氮呈显著负相关(P <0.05),优势种为Penicillium commune;理化数据显示,在17个样地中酸性土壤占比较大,且各样地理化因子间均存在显著性差异(P <0.05);铵态氮、有效磷是影响察隅县水稻根际丝状真菌多样性的主要理化因子.上述结果表明西藏林芝市察隅县水稻根际土壤丝状真菌资源比较丰富并存在明显的空间异质性,土壤理化因子对丝状真菌群落结构具有一定影响.To investigate the community structure of filamentous fungi in rice rhizosphere soil in Chayu County,Nyingchi City, Tibet, China, and to analyze the influence of environmental factors on the filamentous fungal diversity, filamentous fungi were isolated by dilution and plating and identified by sequencing the ITS domains of the rRNA gene and morphological characterization. The physical and chemical indices of soil were determined to analyze the correlations between filamentous fungal diversity and environmental factors. In total, 221filamentous fungal strains were isolated from rice rhizosphere soil in Chayu County;these strains belonged to 33 species of 17 genera. Penicillium was the dominant genus, comprising 10 species. Further, NH-N had a significant negative association with Penicillium. Penicillium commune was the dominant species. Acidic soil accounted for a large proportion, and significant differences were observed among environmental factors in 17sample plots(P < 0.05). Also, NH-N, and AP are important factors that affect the diversity of filamentous fungi in the rice rhizosphere soil. The filamentous fungal community from the rice rhizosphere soil in Chayu County indicated high species richness. The filamentous fungal community structure was different at different sites, and environmental factors had a certain influence on the community structure of the filamentous fungi.
