摘  要:目的研究富血小板血浆(platelet-rich plasm,PRP)对股骨头坏死进程的影响。方法30只SD(Sprague-Dawley)大鼠,随机分为3组。造模组:激素+内毒素法建立股骨头坏死模型。PRP组:造模成功后,股骨头内注射PRP,每周1次,连续4周。对照组:正常饲养。4周后处死,HE染色观察骨小梁结构;电镜下观察细胞核、细胞器改变;TUNEL染色统计骨细胞凋亡率。结果HE染色:造模组骨小梁变细,结构紊乱;PRP组骨小梁结构部分恢复;对照组骨小梁结构正常。电镜观察:造模组细胞核受压,细胞器、线粒体萎缩;PRP组骨细胞接近正常,核膜清晰,细胞表面可见微绒毛;对照组骨细胞正常。TUNEL染色统计骨细胞凋亡率:造模组对比PRP组,细胞凋亡率显著升高;造模组对比对照组,细胞凋亡率显著升高;PRP组对比对照组,细胞凋亡率升高。结论PRP可以延缓股骨头坏死进程。Objective To study the effect of platelet-rich plasma(PRP)on the necrosis of the femoral head.Methods 30 SD(Sprague-Dawley)rats were randomly divided into 3 groups.Methods The femoral head necrosis model was established by the hormone+LPS method.PRP group:After successful mold making,PRP was injected into the femoral head once a week for 4 consecutive weeks.Control group:normal feeding.a tatol of SD rats were killed after 4 weeks,and trabecular structure was observed by HE staining;nuclei and organelles were changed by electron microscopy;TUNEL staining.Results HE staining:molded trabecular structure thinning and disorder;trabecular structure in PRP group;control group was normal.Electron microscopy:module cell nucleus compression,organelles and mitochondria atrophy;PRP bone cells near normal,clear nuclear membrane,visible microvilli on the cell surface;control bone cells were normal.Statistical apoptotic rate by TUNEL staining:apoptosis rate increased in PRP.Conclusion PRP can delay the process of necrosis of the femoral head.
