摘 要:研究组蛋白H3K27乙酰化(H3K27ac)对三基序蛋白家族11(TRIM11)的表达及结肠癌细胞增殖的影响。用RT-PCR分析TRIM11 mRNA的表达,蛋白印迹分析TRIM11及H3K27ac的表达水平;用乙酰化酶及去乙酰化酶抑制剂处理细胞,分析H3K27ac水平与TRIM11基因表达水平之间的关系。通过敲低或过表达TRIM11,用CCK8、细胞集落形成实验和Edu检测TRIM11对细胞增殖能力的影响。结果表明,TRIM11和H3K27ac在结肠癌组织中高表达,且呈正相关。去乙酰化酶抑制剂处理结肠癌细胞后,H3K27ac的表达明显增强,TRIM11的表达也随之增高;乙酰化酶抑制剂处理结肠癌细胞后,结肠癌细胞H3K27ac的表达显著降低,TRIM11的表达也随之降低。TRIM11高表达可促进结肠癌细胞增殖。TRIM11在结肠癌发生发展中起促进作用,有望成为结肠癌潜在标志物。This study was to examine the effect of H3K27 acetylation(H3K27ac) on the expression level of tripartite motif 11(TRIM11) in colon cancer and proliferation of colon cancer cells. RT-PCR and Western blot were used to detect gene transcription and protein expression level. Acetylase and deacetylase inhibitors were used to treat colon cancer cell to analyze the relationship between H3K27ac levels and TRIM11 levels. CCK8, colony formation test and Edu methods were used to study the effect of TRIM11on cell proliferation through knockdown or overexpression of TRIM11. The results showed that the TRIM11 and H3K27ac were highly expressed in colon cancer tissues. The expression of the TRIM11 and H3K27ac was correspondingly decreased or increased in colon cancer cells treated with histone acetylase inhibitor or deacetylase(HDAC) inhibitor. This suggests that there is significant correlation between TRIM11 and H3K27ac. The overexpression or knockdown of TRIM11 could promote or inhibit the proliferation of colon cancer cells. TRIM11 plays an role in promoting the occurrence and development of colon cancer, and is expected to become a potential marker of colon cancer.
