摘　　要：目的:探索皖北地区孕妇人群中IgG3同种异型p.Arg435His(p.Arg435His-IgG3)的阳性率,为后续进一步研究p.Arg435His-IgG3与胎儿和新生儿溶血病的发生发展奠定理论基础。方法:提取177例孕妇血液样本DNA,设计特定引物,采用PCR技术对IgG3重链恒定区CH2和CH3的基因片段进行扩增,再通过Sanger测序法对扩增产物进行基因测序。将测序结果与参考序列AJ390235(IMGT基因库登录号)进行比对,以384位氨基酸密码子突变作为实验内参,查找435位氨基酸密码子是否发生突变并计算出p.Arg435His-IgG3的阳性率。结果:177例孕妇血样中有3例无效样本,在其余174例有效样本中检测到一例为IgG3同种异型p.Arg435His(g.1053927G>A),阳性率为0.57%;通过比对384位氨基酸密码子测序结果提示,在174例样本中有纯合子AGC 30例、AAT 61例,杂合子AG/AC/T 83例。结论:皖北地区孕妇人群中存在p.Arg435His-IgG3同种异型,其阳性率与继往研究相比偏低。Objective:To explore the positive rate of IgG3 allotype p.Arg435 His(p.Arg435 His-IgG3) of pregnant women in Northern Anhui, and to lay a theoretical foundation for further studies on relationship between p.Arg435 His-IgG3 and occurrence and severity of fetal and neonatal hemolytic diseases.Methods:DNA was extracted from blood samples of 177 pregnant women, specific primers were designed, and the gene fragments of CH2 and CH3 of IgG3 heavy chain constant region were amplified by PCR technology, and then the amplified products were sequenced by Sanger sequencing method. The sequencing results were compared with the reference sequence AJ390235(IMGT gene bank accession number) to find out whether the mutation occurred at position 435 and calculate the positive rate of p.arg435 his-IgG3, with the mutation of the amino acid codon at position 384 being used as the experimental internal reference.Results:There were three invalid samples of the 177 pregnant women’s blood samples, and IgG3 allotype p.Arg435 His(g.1053927 G>A) was found in only one of the remaining 174 valid samples, and the positive rate was 0.57%.The results of the 384-position amino acid codon sequencing showed that there were 30 AGC homozygotes, 61 AAT homozygotes and 83 AG/AC/T heterozygotes in the174 samples.Conclusion:P.Arg435 His-IgG3 was detected in pregnant women in Northern Anhui Province, and its positive rate was low compared to related studies.
