P-选择素靶向碳化铁纳米颗粒的易损斑块磁共振成像研究

摘  要:目的基于Fe_(5)C_(2)碳化铁纳米颗粒构建具有P-选择素靶向功能的磁共振对比剂,用于易损斑块的磁共振成像(MRI)识别。方法用溴化胺诱导法合成Fe_(5)C_(2)碳化铁纳米颗粒,用氨基PEG2000-生物素进行包裹后与Dylight649亲和素孵育获得亲和素-生物素碳化铁纳米颗粒。再与生物素化P-选择素抗体孵育获得P-选择素抗体碳化铁纳米颗粒。给ApoE-/-动脉粥样硬化小鼠尾静脉注射PBS(对照组)、亲和素-生物素碳化铁纳米颗粒(亲和素组)和P-选择素抗体碳化铁纳米颗粒(P-选择素组)24 h后用7.0T小动物MRI扫描获取T1 Flash和T2 TurboRARE序列图像。使用ImageJ分析MRI图像获取斑块信号强度,计算斑块与邻近肌肉信号的相对信号强度(rSI)。结果对照组、亲和素组和P-选择素组的rSI分别为(1.83±0.18,1.41±0.31,3.69±0.48,P=0.002)。P-选择素组与对照组(P=0.004)和亲和素组(P=0.001)间差异有统计学意义,对照组和亲和素组间差异无统计学意义(P=0.173)。结论P-选择素修饰的Fe_(5)C_(2)碳化铁纳米颗粒可以有效靶向易损斑块,提高图像对比度。【Objective】To construct magnetic resonance contrast agents with P-selectin targeting function based on Fe_(5)C_(2) iron carbide nanoparticles for magnetic resonance imaging(MRI)to identify vulnerable plaques.【Methods】Fe_(5)C_(2) iron carbide nanoparticles were synthesized by the amine bromide induction method,wrapped with amine-PEG2000-biotin and incubated with Dylight649 streptavidin to obtain streptavidin-biotin iron carbide nanoparticles.Then incubated with biotinylated P-selectin antibody to obtain P-selectin antibody iron carbide nanoparticles.ApoE-/-atherosclerotic mice were given tail vein injections of PBS(control group),streptavidin-biotin iron carbide nanoparticles(streptavidin group)and P-selectin antibody iron carbide nanoparticles(Pselectin group),then T1 FLASH and T2 TurboRARE sequence images were obtained using 7.0T small animal MRI scans after 24 hours.MRI images were analyzed using ImageJ to acquire plaque signal intensity and calculate the relative signal intensity(rSI)of the plaque to the adjacent muscle signal.【Results】The rSI values were 1.83±0.18(control group),1.41±0.31(streptavidin group)and 3.69±0.48(P-selectin group)(P=0.002).The rSI value of P-selectin group was higher than control group(P=0.004)and streptavidin group(P=0.001).While the differences between control group and streptavidin group were not statistically significant(P=0.173).【Conclusion】P-selectin-modified Fe_(5)C_(2) iron carbide nanoparticles can effectively target vulnerable plaques and improve image contrast.
