摘 要:目的观察针刺对抽动障碍模型大鼠脑中儿茶酚胺类降解酶COMT、MAO-B和多巴胺受体D1、D2表达的影响。方法实验大鼠造模成功后随机分为模型组、针刺治疗组、对照组,同时设立空白组正常饲养,每组10只。治疗组予针刺治疗,对照组予氟哌啶醇片灌胃,空白组与模型组给予等体积生理盐水灌胃,治疗4周。治疗结束后取脑组织,检测COMT、MAO-B、DRD1、DRD2的mRNA表达量。结果治疗4周后,与模型组和对照组比较,针刺组大鼠脑中COMT、MAO-B的mRNA表达量均明显升高(P<0.05);与模型组比较,针刺组大鼠脑中DRD1的mRNA表达量明显降低,DRD2的mRNA表达量明显升高(P<0.05)。结论针刺治疗能通过影响IDPN模型大鼠脑中COMT、MAO-B、DRD1、DRD2的表达水平达到治疗抽动障碍的作用。Objective To observe the effects of acupuncture on the expression levels of catecholamine-degrading enzymes COMT,MAO-B and dopamine receptors D1 and D2 in brain of tic disorder(TD)model rats.Methods After successful modeling,experimental rats were randomly divided into model group,acupuncture group and control group.At the same time,blank group was set up to feed normally,with 10 rats in each group.The treatment group was treated with acupuncture,the control group was given haloperidol by gavage,and the blank group and model group were given equal volume pure water for 4 weeks.After treatment,brain tissues were taken to detect the mRNA expression levels of COMT,MAO-B,DRD1 and DRD2.Results After 4 weeks of treatment,compared with model group and control group,the mRNA expression levels of COMT and MAO-B in brain of acupuncture group were significantly increased(P<0.05).Compared with model group,the mRNA expression level of DRD1 was significantly decreased,while the mRNA expression level of DRD2 was significantly increased(P<0.05).Conclusion Acupuncture therapy can treat tic disorder by affecting the contents of COMT,MAO-B,DRD1 and DRD2 in the brain of IDPN model rats.
