摘  要:目的:探讨黄芪多糖对过敏性鼻炎大鼠Treg/Th17细胞免疫平衡和炎症因子的影响。方法:将50只SD大鼠分为空白组、模型组、氯雷他定组和黄芪多糖低剂量组、黄芪多糖高剂量组,每组10只。应用卵白蛋白+氢氧化铝粉建立过敏性鼻炎基础致敏模型。观察大鼠基础行为学改变,观察HE染色的病理情况,采用ELISA法检测大鼠血清IgE、白介素-10(IL-10)、白介素-17(IL-17)水平,流式细胞术检测血清Th17/Treg分布。结果:与模型组比较,氯雷他定组、黄芪多糖低剂量组、黄芪多糖高剂量组鼻黏膜组织上皮组织较为完整,有少量的炎性浸润;模型组行为学积分和血清IgE、IL-17、Th17水平显著升高(均P<0.05),IL-10、Treg水平显著降低(均P<0.05)。与模型组比较,黄芪多糖高剂量组大鼠IL-10、Treg水平显著升高(均P<0.05),行为学积分和血清IgE、IL-17、Th17水平显著降低(均P<0.05)。结论:黄芪多糖可有效缓解过敏性鼻炎大鼠模型的症状,机制可能与提高免疫功能、抑制炎症反应有关。Objective:To investigate the effect of astragalus polysaccharide on immune balance and inflammatory factors of Treg/Th17 cells in a rat model of allergic rhinitis.Methods:Fifty SD rats were divided into blank group,model group,loratadine group and low and high dose groups of astragalus polysaccharide,10 rats in each group.The basic sensitization model of allergic rhinitis was established by using ovalbumin+aluminum hydroxide powder.The changes of the basic behavior of the rats were observed,the serum levels of IgE,IL-10 and IL-17 were detected by ELISA,and the distribution of serum Th17/Treg was detected by flow cytometry.Results:Compared with the blank group,the behavioral scores and serum IgE,IL-17 and Th17 levels in the model group were significantly increased(all P<0.05),while the IL-10 and Treg levels were significantly decreased(all P<0.05).Compared with the model group,the levels of IL-10 and Treg in the high-dose astragalus polysaccharide group were significantly increased(all P<0.05),and the behavioral scores and serum IgE,IL-17,and Th17 levels were significantly decreased(all P<0.05).Conclusion:Astragalus polysaccharide can effectively relieve the symptoms of allergic rhinitis rat model,and the mechanism may be related to improving immune function and inhibiting inflammatory response.
