摘 要:目的观察益肾通络方对聚苯乙烯微塑料(PS-MPs)致大鼠精子DNA损伤的修复作用。方法随机将24只雄性大鼠分为空白组(灌胃生理盐水)、模型组(按10mg/kg灌胃聚苯乙烯微球)、YSTL组(在模型组的基础上按照10.44g/kg灌胃益肾通络方水煎液)。8周后采用精子染色质结构分析法(SCSA)测定各组大鼠精子DNA碎片指数(DFI),采用ELISA法检测各组大鼠睾丸SOD、MDA、ATP、NO及血清FSH、LH、T水平。结果①与空白组相比,模型组大鼠精子DFI显著升高(P<0.01),SOD、ATP显著降低,MDA、NO显著升高,血清LH、T显著降低(P<0.05)。②YSTL组与模型组相比,DFI明显降低(P<0.05),MDA、ATP明显恢复(P<0.05)。结论微塑料可造成大鼠精子DNA氧化损伤,性激素水平受到抑制。益肾方可修复由微塑料引起的大鼠精子DNA损伤,作用机制可能与抗氧化有关。Objective To observe the repairing effect of Yishen Tongluo formula on DNA damage of spermatozoa caused by polystyrene microplastics(PS-MPs)in rats.Methods Twenty-four male rats were randomly divided into the control group,model group and YSTL group.The model group was gavaged with polystyrene microspheres at 10mg/kg,the YSTL group was gavaged with Yishen Tongluo decoction at 10.44 g/kg on the basis of the model group,and the control group were gavaged with the same amount of saline.8 weeks later,the sperm DNA fragmentation index(DFI)of each group was measured by sperm chromatin structure analysis(SCSA),and the testicular SOD,MDA,ATP,NO,and SOD of each group were measured by ELISA.MDA,ATP,NO and serum FSH,LH and T levels were measured by ELISA.Results 1.Compared with the control group,DFI of sperm in the model group was significantly increased(P<0.01),SOD and ATP were significantly decreased,MDA and NO were significantly increased,and serum LH and T were significantly decreased in the model group(P<0.05).2.The DFI was significantly decreased(P<0.05),and MDA and ATP were restored(P<0.05)in the YSTL group compared with the model group.Conclusion Microplastics can cause oxidative damage to sperm DNA and suppression of sex hormone levels in rats.The mechanism of action of YSTL in repairing the DNA damage of rat sperm caused by microplastics may be related to antioxidants.
