摘  要:目的探索四逆汤对慢性心力衰竭大鼠心肌细胞凋亡的影响。方法将24只大鼠随机分为空白组、模型组、美托洛尔组、四逆汤组。除空白组外,其余大鼠利用腹腔注射阿霉素建立慢性心力衰竭模型。灌胃给药28天后,利用彩超仪检测大鼠心功能;利用ELISA法检测心肌组织Bax、Bcl-2、Caspase-3、Caspase-9的浓度改变;Western blot观察心肌组织Bax、Bcl-2、Caspase-3、Caspase-9蛋白含量;Real-time PCR法检测心肌组织的Bax、Bcl-2、Caspase-3、Caspase-9 mRNA相对表达量情况。结果与空白组相比,模型组大鼠的左心室射血分数(EF)、左心室短轴缩短率(FS)显著降低(P<0.01),BAX、Caspase-3、Caspase-9的浓度、mRNA含量以及蛋白表达水平显著上升(P<0.01),Bcl-2的浓度、mRNA含量以及蛋白表达水平显著降低(P<0.01);与模型组相比,四逆汤EF、FS值显著升高(P<0.01),BAX、Caspase-3、Caspase-9的浓度、mRNA含量以及蛋白表达水平显著降低(P<0.01),Bcl-2的浓度、mRNA含量以及蛋白表达水平显著提高(P<0.01)。结论四逆汤可通过调控心肌细胞凋亡来改善慢性心力衰竭大鼠的心功能。Objective To explore the effect of Sini Decoction on cardiomyocyte apoptosis in rats with chronic heart failure.Methods Twenty-four rats were randomly divided into blank group,model group,metoprolol group,and Sini decoction group.Except for the blank group,the other rats used adriamycin intraperitoneally to establish chronic heart failure models.After 28 days of intragastric administration,the rat heart function was detected by color Doppler ultrasound;the concentration of Bax,Bcl-2,Caspase-3,and Caspase-9 in myocardial tissue were detected by ELISA;Caspase-3 and Caspase-9 protein content;Real-time PCR method was used to detect the relative expression of Bax,Bcl-2,Caspase-3 and Caspase-9 mRNA in myocardial tissue.Results Compared with the blank group,the left ventricular ejection fraction(EF)and left ventricular short axis shortening rate(FS)of the model group were significantly reduced(P<0.01),and the concentrations of BAX,Caspase-3,and Caspase-9,MRNA content and protein expression level increased significantly(P<0.01),Bcl-2 concentration,mRNA content,and protein expression level significantly decreased(P<0.01);compared with the model group,Sini Decoction EF and FS values significantly increased High(P<0.01),the concentration,mRNA content and protein expression level of BAX,Caspase-3,Caspase-9 were significantly reduced(P<0.01),and the concentration of Bcl-2,mRNA content and protein expression level were significantly increased(P<0.01).Conclusion Sini Decoction can improve the cardiac function of rats with chronic heart failure by regulating the apoptosis of cardiomyocytes.
