摘  要:目的分析早期生长反应因子-1(early growth response 1,Egr1)对增生性瘢痕成纤维细胞生长的影响。方法选取2018年1月至2019年1月于北部战区总医院烧伤整形科就诊的12例增生性瘢痕患者的瘢痕组织作为研究对象,选取同期就诊的12例外伤皮瓣术后患者剩余皮肤组织作为对照组。Western blot实验检测组织中Egr1表达情况。提取增生性瘢痕成纤维细胞,分别转染si-NC和si-Egr1,通过MTT实验检测0、1、2、3和4 d时细胞增殖情况,细胞周期实验检测Egr1对细胞周期影响,细胞凋亡实验检测Egr1对细胞凋亡影响。Western blot实验检测Egr1对于Cyclin D1和p21蛋白表达影响。结果增生性瘢痕组织中Egr1表达水平高于对照组。MTT实验检测结果发现,与转染si-NC相比,si-Egr1转染后,抑制增生性瘢痕成纤维细胞的增殖(P<0.05)。抑制增生性瘢痕成纤维细胞的细胞周期G1/S期转导(P<0.05),显著促进了增生性瘢痕成纤维细胞凋亡(P<0.05)。si-Egr1组增生性瘢痕成纤维细胞中Egr1、Cyclin D1和p21表达下调。结论Egr1在增生性瘢痕组织中表达上调,有望为瘢痕治疗寻求新的治疗靶点。Objective To analyze the effect of early growth response factor-1(Egr1)on the growth of hypertrophic scar fibroblasts.Methods From January 2018 to January 2019,12 patients with hypertrophic scar in the Department of Burn and Plastic Surgery of General Hospital of Northern Theater Command were selected as research subjects.The remaining skin tissues of skin flap in 12 patients underwent post-traumatic reconstruction in the same period were selected as the control group.The expression of Egr1 was detected by Western blot.Hypertrophic scar fibroblasts were extracted and transfected with si-NC and si-Egr1,respectively.The proliferation of fibroblasts at 0,1,2,3 and 4 days was detected by MTT assay,the effect of Egr1 on cell cycle was detected by cell cycle assay and the effect of Egr1 on apoptosis was detected by cell apoptosis assay.The effect of Egr1 on the expression of cyclin D1 and p21 was detected by Western blot.Results The expression level of Egr1 in hypertrophic scar was higher than that in the control group.MTT assay showed that si-Egr1 transfection inhibited the proliferation of hypertrophic scar fibroblasts compared with si-NC transfection(P<0.05).Compared with control group(si-NC transfection),si-Egr1 transfection inhibited the G1/S-phase transition of hypertrophic scar fibroblasts(P<0.05)and significantly promoted the apoptosis of hypertrophic scar fibroblasts(P<0.05).The expression of Egr1,cyclin D1 and p21 were down regulated in hypertrophic scar fibroblasts in si-Egr1 group.Conclusion The expression of Egr1 is up-regulated in hypertrophic scar tissue,which is expected to seek a new therapeutic target for scar treatment.
