摘 要:为探究不同浓度铅胁迫下的鸡腿菇菌丝生长状态,了解鸡腿菇菌丝对铅的耐受性及富集情况,通过不同Pb^(2+)浓度处理鸡腿菇菌丝,测定菌丝生长速率,铅含量、丙二醛(MDA)含量,超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性。结果:Pb^(2+)浓度较低(≤10 mg/L)时,能促进鸡腿菇菌丝生长,Pb^(2+)浓度较高(15~25 mg/L)时会抑制菌丝生长,浓度越高,抑制作用越明显;菌丝体铅含量与培养基中的Pb^(2+)浓度呈正相关,在Pb^(2+)浓度25 mg/L时达到最大值1.45 mg/kg,超过国家限量标准;随着Pb^(2+)浓度的增大,菌丝体MDA含量及抗氧化酶活性均呈先升高后降低的变化规律,一定程度解释了鸡腿菇菌丝体在铅胁迫下的防御反应。相关性分析结果表明,菌丝生长速率、铅离子富集量与多个抗氧化酶指标之间有极显著相关性。The objective of the present study is to study the effects of lead stress on the growth of Coprinus comatus mycelium and to understand the tolerance and accumulation of lead in Coprinus comatus mycelium. Mycelium of Coprinus comatus was treated with different concentrations of Pb^(2+), the mycelial growth rate, lead content,malondialdehyde(MDA) content, superoxide dismutase(SOD) activity, peroxidase(POD) activity and catalase(CAT)activity of Coprinus comatus mycelium were determined. The results showed that when the concentration of Pb^(2+) was low, the mycelium growth of Coprinus comatus was promoted, but the mycelium growth rate of Coprinus comatus was inhibited by high concentration of Pb^(2+). The content of lead in the mycelium of Coprinus comatus was positively correlated with the concentration of Pb^(2+) in the culture medium, and reached the maximum at 25 mg/L, which was 1.45mg/kg. Under the treatment of low Pb^(2+) concentration, the concentration of Pb^(2+) in the mycelium of Coprinus comatus was lower than the limit of lead in food, and when the concentration reached 25 mg/L, the enrichment content exceeded the limit of lead in food. The content of MDA and the activity of antioxidant enzymes in the mycelium of Coprinus comatus in liquid medium increased at first and then decreased with the increase of Pb^(2+) concentration. With the increase of Pb^(2+) concentration, the activities of MDA and antioxidant enzymes began to increase. When the concentration was 10 mg/L, the CAT activity of Coprinus comatus mycelium decreased at first, while the MDA content and POD, SOD activity continued to increase;when the Pb^(2+)concentration reached 20 mg/L inch, the mycelium MDA content and POD, SOD activity reached the peak, and then began to decrease, which explained the defense response of Coprinus comatus mycelium under lead stress to some extent. The results of correlation analysis showed that the mycelial growth rate and lead ion concentration were significantly correlated with several antioxidant enzymes.
