摘 要:[目的]探讨柘木总黄酮对白血病K562细胞增殖、凋亡及EGR1/TP53通路的影响。[方法]体外培养K562细胞,阴性对照组细胞不进行处理;阳性对照组用终浓度为10 mg/mL的甲磺酸伊马替尼干预,低、中、高剂量组分别用终浓度为50、100、200 mg/mL的柘木总黄酮干预。48 h后分别采用MTT法和流式细胞术检测各组细胞增殖和凋亡情况,同时采用ELISA法检测各组细胞中IL-6、IL-12、TNF-α水平,Western Blot法检测各组细胞中EGR1、TP53、Wnt、β-Catenin和ERK1/2蛋白水平。[结果]通过生物学数据库http://wwww.hprd.org查找EGR1的相互作用蛋白,发现TP53与EGR1相互作用;与阴性对照组比较,阳性对照组和柘木总黄酮低、中、高剂量细胞增殖率和Wnt、β-Catenin、ERK1/2蛋白水平降低,细胞凋亡率、IL-6、IL-12、TNF-α和EGR1、TP53蛋白水平增加(P<0.05);与阳性对照组比较,各柘木总黄酮剂量组细胞增殖率和Wnt、β-catenin、ERK1/2蛋白水平增加,细胞凋亡率、IL-6、IL-12、TNF-α和EGR1、TP53蛋白水平降低,且随着柘木总黄酮剂量增加,柘木总黄酮各剂量组各指标呈剂量反应关系(P<0.05)。[结论]柘木总黄酮通过激活EGR1/TP53通路,促进K562细胞凋亡,抑制K562细胞增殖。[Objective]To investigate the effect of total flavonoids from Cudrania tricuspidata on proliferation,apoptosis and EGR1/TP53 pathway of leukemia K562 cells.[Method]K562 cells were cultured in vitro,and the negative control group cells were not treated,the positive control group intervened with imatinib mesylate at the final concentration of 10mg/mL,and the low,medium and high-dose groups were intervened with total flavones from Cudrania tricuspidata at the final concentration of 50,100 and 200 mg/ml,respectively.After 48 hours,MTT assay and flow cytometry were used to detect cell proliferation and apoptosis,and ELISA was used to detect IL-6,IL-12 and TNF-α,and Western Blot was used to detect the protein expression of EGR1,TP53,Wnt,β-atenin and ERK1/2.[Result]Through the biological database http://www.hprd.org to search for the interacting protein of EGR1,it was found that TP53 interacted with EGR1.Compared with the negative control group,the cell proliferation rates and Wnt,ERK1/2 protein levels in the positive control group and the low,medium,and high-dose total flavonoids from Cudrania tricuspidata groups were decreased,and cell apoptosis rate,IL-6,IL-12,TNF-αand protein levels of EGR1,TP53 increased(P<0.05).Compared with the positive control group,the cell proliferation rates and Wnt,β-catenin,ERK1/2 protein levels in the low,medium,and high-dose total flavonoids from Cudrania tricuspidata groups were increased,and the apoptosis rate,IL-6,IL-12,TNF-αand EGR1,TP53 protein levels decreased,and with the dose of flavonoids increased,and the indicators of each dose group total flavonoids from Cudrania tricuspidata showed a dose-response relationship(P<0.05).[Conclusion]Total flavonoids from cudrania tricuspidata can promote the apoptosis of K562 cells and inhibit the proliferation of K562 cells by activating EGR1/TP53 pathway.
