摘  要:目的探讨丹参酮ⅡA(TanⅡA)对大鼠创面愈合瘢痕增生的抑制作用。方法取96只大鼠建立创面模型,随机分为模型组及TanⅡA低、中和高剂量组,其余24只为对照组。TanⅡA低、中和高剂量组分别按100、200和300 mg·kg^(-1)的剂量制成10 mL TanⅡA溶液腹腔注射,对照组、模型组腹腔注射DMSO 10 mL,连续注射14 d。比较各组大鼠创面组织瘢痕增生指数(HI)、单位面积的成纤维细胞数量(NA)、创面组织学、转化生长因子β_(1)(TGF-β_(1))、肿瘤坏死因子-α(TNF-α)、白介素-10(IL-10)、粒-单系祖细胞集落刺激因子(GM-CSF)mRNA及蛋白表达的差异。结果HE染色显示,注射14 d后对照组创面组织上皮完整、无炎性细胞浸润,模型组可见成纤维细胞(FB)肿胀、排列紊乱,有大量炎性细胞浸润;TanⅡA不同剂量组注射3 d即可见FB增生,高剂量组最明显;注射14 d后,TanⅡA高剂量组FB轻微肿胀,有少量炎性细胞浸润。TanⅡA不同剂量组在各注射时间创面愈合面积、TGF-β_(1)水平、IL-10、GM-CSF mRNA和蛋白水平较模型组升高(P<0.05),注射3、7 d后,HI、NA较模型组升高(P<0.05);愈合时间、TNF-α水平和注射14 d的TanⅡA中、低剂量组HI、NA较模型组降低(P<0.05)。随TanⅡA剂量增加,HI、NA、创面愈合面积、TGF-β_(1)水平、IL-10、GM-CSF mRNA和蛋白水平增加,创面愈合时间缩短,TNF-α水平降低(P<0.05)。结论TanⅡA对大鼠创面修复中增生性瘢痕的产生有较明显的抑制作用,其机制可能与IL-10、GM-CSF mRNA和蛋白的表达升高有关。Objective To investigate the inhibition effect of tanshinoneⅡA(TanⅡA)on hypertrophic scar of rats.Methods 96 rats,after scar model establishment,were randomly divided into model group,TanⅡA low,medium and high dose groups and the rest 24 rats were grouped as a control.The TanⅡA low,medium and high dose groups were injected with 10 mL of 100,200 and 300 mg·kg^(-1) TanⅡA solution,and 10 mL DMSO were injected in the control and model groups,consecutively injected for 14 days.The scar hyperplasia index(HI),number of fibroblasts in unit area(NA),scar histopathology,and differences in transforming growth factor-beta1(TGF-β_(1)),tumor necrosis factor-α(TNF-α)level,interleukin-10(IL-10)and granulocyte-macrophage colon stimulating factor(GM-CSF)mRNA and protein expression were compared among the groups.Results The results of HE staining showed the control group wound tissue was intact and without inflammatory cell infiltration.The model group after injection 14 days showed fibroblasts(FB)were swollen and disordered,and a large number of inflammatory cell infiltration;FB proliferation was observed in TanⅡA different dose groups after injected 3 days,with the most significant increased in the TanⅡA high dose group.The FB of TanⅡA high dose group after injection 14 days,was still slightly swollen with a small infiltration of inflammatory cells.Compared with the model group,the healing scar area,TGF-β_(1),the levels of IL-10,GM-CSF mRNA and protein expression in different TanⅡA dose groups were significantly increased(P<0.05).HI and NA after injection 3 days and 7 days in different TanⅡA dose groups were significantly increased(P<0.05),while the wound healing time,TNF-α,and the HI,NA of injection after 14 days in TanⅡA medium and low dose group were significantly decreased(P<0.05).With the TanⅡA dose increasing,the HI,NA,healing scar area,TGF-β_(1),IL-10,GM-CSF mRNA,and protein levels of IL-10,GM-CSF were increased and the wound healing time and TNF-αlevels were decreased(P<0.05).Conclusion Tan
