摘  要:蔗糖合成酶参与植物非生物胁迫,且在调控植物代谢方面起重要作用。为了明确蓝果忍冬蔗糖合成酶对干旱胁迫的响应情况,从蓝果忍冬cDNA中克隆一个蔗糖合成酶基因,命名为LcSUS1。该基因cDNA全长1206 bp,编码401个氨基酸,分子量为45.78 kD,理论等电点为5.93。LcSUS1的N端有一个明显的跨膜区域。LcSUS1的氨基酸序列与可可等植物的SUS1序列的同源性较高。对LcSUS1基因在蓝果忍冬不同组织部位的表达情况研究发现,LcSUS1基因在蓝果忍冬茎中表达量最高。RNA-Seq和实时定量PCR数据表明该基因在干旱胁迫下表达量上升,说明LcSUS1可能在响应干旱胁迫过程中发挥重要作用。Sucrose synthase is involved in plant abiotic stress and plays an important role in the regulation of metabolism for plants.In order to identify how the sucrose synthase will response to drought stress,a gene of sucrose synthase,LcSUS1,which was cloned from the c DNA of Lonicera caerulea.The gene was 1206 bp in length and was predicted to encode a polypeptide of 401 amino acids.The estimated molecular weight and isoelectric point of the putative protein were 45.78 kD and 5.93,respectively.The N-terminal of LcSUS1 has a distinct transmembrane domain.The amino acid sequence of LcSUS1 shares a relatively high homology with the one in plants such as coco.After,studying on the expression of LcSUS1 in different tissues and organs,it was found that the expression level of LcSUS1 gene was the highest in the stem of Lonicera caerulea.RNA-Seq and real-time PCR have proved that the expression of this gene increased under drought stress,indicating that LcSUS1 may play an important role in response to drought stress.
