摘 要:CLE(CLAVATA3/Embryo surrounding region-related)多肽对植物根的生长发育具有重要调控作用。本研究采用生物信息学方法对人参CLE基因进行鉴定,对其基序进行了比对、偏好性分析,并采用全长进行了系统进化分析,对人参CLE基序在根的生长发育功能进行初步研究。结果表明,从人参基因组及转录组对应的蛋白库中鉴定出15种人参CLE多肽序列,对应21个基因,命名为PgCLE+数字,其中11种多肽序列为首次报道;多序列比对及偏好性分析表明,人参PgCLE与拟南芥CLE的同源多肽较为一致,但是部分人参CLE在第3位氨基酸上表现出了对A氨基酸的偏好性;系统进化树表明,PgCLE1、PgCLE3、PgCLE5、PgCLE6、PgCLE25a/b、PgCLE26、PgCLE27、PgCLE45a/b/c/d聚类在一起,PgCLE9、PgCLE12、PgCLE14a/b、PgCLE16、PgCLE20、PgCLE41、PgCLE44a/b聚类在一起,均同拟南芥对应的CLE亲缘关系较近;采用人参不定根系统研究发现,PgCLE9、PgCLE14、PgCLE16、PgCLE20、PgCLE26和PgCLE44对人参不定根分支数存在抑制作用,而PgCLE5具有促进作用;采用拟南芥初生根系统研究发现,PgCLE1、PgCLE9、PgCLE14、PgCLE16、PgCLE20和PgCLE26抑制初生根生长显著,而拟南芥bam双突变体则对以上6种PgCLE不敏感,表明受体激酶BAM可能为这6种PgCLE的受体蛋白。本研究对人参的CLE家族序列特征和系统进化关系进行分析,初步探究了人参PgCLE对根生长发育的功能,为人参根型的调控提供理论依据。CLE polypeptide plays an important role in regulating the growth and development of plant roots.In this study,the CLE genes were identified in Panax ginseng by bioinformatics approaches.CLE motifs were used for comparison and preference analysis while the full-length for phylogenetic analysis.The growth and development function of CLE motif in first-reported ginseng roots was preliminarily studied.The results showed that 15 ginseng CLE polypeptides were identified from the corresponding protein library of ginseng genome and transcriptome,corresponding to 21 genes,named PgCLE+number,11 of which were reported for the first time.Multiple sequence alignment and preference analysis showed that the homologous polypeptides of ginseng PgCLE and Ara bidopsis CLE were consistent,but some ginseng CLE showed preference for the amino acid of A in the third place.Phylogenetic tree showed that PgCLE1,PgCLE3,PgCLE5,PgCLE6,PgCLE25a/b,PgCLE26,PgCLE27,PgCLE45a/b/c/d were clustered together,and PgCLE9,PgCLE12,PgCLE14a/b,PgCLE16,PgCLE20,PgCLE41,PgCLE44a/b were clustered together,which were closely related to the corresponding CLE of Arabidopsis thaliana.PgCLE9,PgCLE14,PgCLE16,PgCLE20,PgCLE26 and PgCLE44 inhibited the number of branches of ginseng adventitious roots,while PgCLE5 promoted the number of branches.PgCLE1,PgCLE9,PgCLE14,PgCLE16,PgCLE20 and PgCLE26 significantly inhibited the growth of primary roots in Arabidopsis thaliana.Interestingly,bam double mutants were insensitive to PgCLE1,PgCLE9,PgCLE14,PgCLE16,PgCLE20 and PgCLE26,which suggested that BAM might be the putative receptor of the above six PgCLEs.In this study,the sequence characteristics and phylogenetic relationship of ginseng CLE family were analyzed,and the function of ginseng PgCLE on root growth and development was preliminarily explored,which will provide theoretical basis for the regulation of ginseng root type.
