萝芙木内生真菌Colletotrichum acutatum的次级代谢产物研究

摘　　要：目的 对萝芙木内生真菌Colletotrichum acutatum中抗肿瘤活性次级代谢产物进行研究。方法 结合LC-MS分析与MTT活性检测方法,采用反相ODS、反复硅胶柱色谱及高效液相色谱等方法对活性次级代谢物进行分离纯化,并根据理化性质与波谱数据鉴定化合物结构。结果与结论从萝芙木内生真菌总次级代谢物中分离得到11个化合物,分别鉴定为alternariol 4-methl ether(1)、fusaproliferin(2)、对苯二甲酸二丁酯(3)、nectriapyrone(4)、heptadec-(9Z)-enoic acid(5)、bis-2-ethylhexyl-benzene-1,2-dicarboxylate(6)、psoralester(7)、1-linoleoylglycerol(8)、9-octadecenoic acid-2′,3′-dihydroxy propyl ester(9)、rigidiusculamide C(10)、sambutoxin(11)。其中除化合物4以外,其他化合物均为首次从内生真菌Colletotrichum属中分离得到,化合物3、7、8、11对肺癌细胞株A549和宫颈癌细胞株HeLa具有中等细胞毒性(IC_(50)≤50μmol·L^(-1))。Screened out an antitumor endophytic fungus Colletotrichum acutatum based on LC-MS analysis.Eleven secondary metabolites were isolated and purified by using ODS flash column,thin layer chromatography,silica gel column chromatography and high performance liquid chromatography.The compounds were identified as alternariol 4-methl ether(1),fusproliferin(2),dibutylterephthalate(3),nectriapyrone(4),heptadec-(9Z)-enoic acid(5),bis-2-ethylhexyl-benzene-1,2-dicarboxylate(6),psoralester(7),1-linoleoylglycerol(8),9-octadecenoic acid-2′,3′-dihydroxy propyl ester(9),rigidius-culamide C(10),sambutoxin(11)by extensive spectroscopic analysis.Ten compounds were isolated from the Colletotrichum for the first time except for 4.Compounds 3,7,8,and 11 showed moderate cytotoxic activities against A549 and HeLa cell with the IC50 values of less than 50μmol·L^(-1).
