摘  要:目的观察中医辟谷养生术对肠道菌群的调节作用。方法共计23名受试者进行5~7天辟谷,同时练习养生功法。分别于辟谷前3天、辟谷后3天及2个月采集受试者粪便样本,对样本进行DNA抽提和检测、16S r DNA序列扩增、Mi Seq测序和数据分析。结果23名受试者中18名完成了辟谷干预及3次粪便样本采集。与辟谷前比较,辟谷后2个月拟杆菌丰度在门、纲、目水平均明显增多(P<0.05),厚壁菌门的毛螺菌科丰度明显减少(P<0.05),厚壁菌门/拟杆菌门比例由辟谷前的0.65±0.67,降至辟谷后0.43±0.25,以及辟谷后2个月的0.40±0.29。辟谷后2个月,“代谢”“糖合成及代谢”“非基本氨基酸代谢”“代谢性疾病”功能菌群丰度均明显增高(P<0.05,P<0.01),“信息处理”“膜转运”功能菌群丰度明显降低(P<0.05)。结论辟谷可改变肠道菌群结构与相关功能。Objective To observe the intervention effect of Bigu on gut microbiota.Methods Totally 23 subjects accepted Bigu and health exercises for 5~7 days.The fecal samples were collected 3 days before,3 days and 2 months after the Bigu intervention.DNA extraction and detection,16S r DNA sequence amplification,Mi Seq sequencing,and data analysis were performed in fecal samples.Results Totally 18 subjects finished the Bigu intervention and fecal samples collection for 3 times.Compared with before Bigu intervention,phylum Bacteroidetes,class Bacteroidia and order Bacteroidales increased(P<0.05),family Lachnospiraceae of phylum Firmicutes decreased after 2 months intervention(P<0.05).The ratio of Firmicutes/Bacteroidetes decreased from 0.65±0.67(3 days before intervention)to 0.43±0.25(3 days after intervention),and 0.40±0.29(2 months after intervention).The microbial gene function related to metabolism,glucose synthesis and metabolism,nonessential amino acid metabolism,metabolic diseases increased(P<0.05,P<0.01),while the microbial gene function related to information processing and membrane transport decreased 2 months after intervention(P<0.05).Conclusion Bigu can change bacterial compositions and related function.
