摘 要:目的:观察三仁汤各提取部位对高脂饮食诱导的非酒精性脂肪性肝病(non-alcoholic fatty liver disease, NAFLD)小鼠的作用。方法:使用水提醇沉、石油醚萃取技术获得三仁汤提取部位1,2,3,以三仁汤整方和过氧化物酶体增殖物激活受体α/γ激动剂Saroglitazar作为对照药物。将小鼠分为正常组,模型组,三仁汤提取部位1,2,3组,三仁汤对照组及Saroglitazar对照组,每组9只。正常组予对照饮食,其余各组予高脂饮食,第13周开始灌胃给药,16周末取材。观测小鼠体质量、进食量、空腹血糖、空腹血清胰岛素、胰岛素抵抗指数、血清谷丙转氨酶(alanine aminotransferase, ALT)、肝脏病理、肝脏甘油三酯(triglyceride, TG)及肝脏γ-谷氨酰转移酶(γ-glutamyl transferase, GGT)。结果:三仁汤各提取部位均显著降低NAFLD小鼠体质量和空腹血清胰岛素,改善肝脏病理、肝脏TG和GGT。提取部位2、3还可显著降低NAFLD小鼠空腹血糖和胰岛素抵抗指数。提取部位1、2还可显著降低NAFLD小鼠血清ALT。结论:三仁汤各提取部位均不同程度改善高脂饮食诱导的小鼠NAFLD,其中提取部位2的作用最接近整方。OBJECTIVE To evaluate the effect of different extract fractions of Sanren Tang on nonalcoholic fatty liver disease(NAFLD mice)induced by high-fat diet.METHODS Sanren Tang extracts 1,2 and 3 were obtained by water decoction, ethanol precipitation and petroleum ether extraction.Sanren Tang and Saroglitazar, the agonist of peroxisome proliferators activated receptors(PPARs)α/γ,were used as positive control.The mice were divided into 7 groups: normal group, model group, Sanren Tang fraction 1 group, Sanren Tang fraction 2 group, Sanren Tang fraction 3 group, Sanren Tang control group and Saroglitazar control group, with 9 mice in each group, which was treated with corresponding drugs from 13^(th) week to the end of the 16^(th) week.The mice in normal group were fed with control diet, while the others were fed with high-fat diet.The body weight, food intake, fasting serum glucose and fasting serum insulin, serum alanine aminotransferase(ALT),hepatic pathology, hepatic triglyceride(TG)and hepatic γ-glutamyl transferase(GGT)were detected.RESULTS Sanren Tang fraction 1,2 and 3 all significantly reduced the body weight and fasting serum insulin of NAFLD mice, improved hepatic histology, hepatic TG and GGT.Furthermore, Sanren Tang fraction 2 and 3 also significantly reduced fasting serum glucose and insulin resistance index of NAFLD mice.Sanren Tang fraction1 and 2 also significantly reduced the serum ALT of NAFLD mice.CONCLUSION All extracted fractions of Sanren Tang can improve NAFLD induced by high-fat diet in mice in varying degrees, and the effect of Sanren Tang fraction 2 was the closest to the whole formula.
