摘  要:目的DNATyper17+28Y复合扩增体系包含16个常染色体短串联重复序列(autosomal short tandem repeat,A-STR)、28个Y染色体短串联重复序列(Y-chromosome short tandem repeat,Y-STR)和1个Amelogenin的复合扩增体系,本研究对该体系进行系列法医学验证后评估其性能指标,为实际应用提供参考。方法本研究从体系灵敏度、种属特异性、混合样本鉴别能力、准确性、重现性、稳定性、耐受性、PCR扩增条件等方面对该体系进行了系统验证。结果该体系灵敏度高,特异性好,具有良好的准确性、重现性、稳定性和耐受性。结论DNATyper17+28 Y复合扩增体系的各项性能指标能满足个体识别、亲缘关系分析、家系排查等检验需求,在法医物证中有应用价值。Objective A DNATyper17+28Y multiplex amplification system contains 16 autosomal short tandem repeat(A-STR),28 Y-chromosome short tandem repeat(Y-STR)and Amelogenin,which is evaluated after a series of forensic verifications to provide a reference for practical applications.Methods In this study,the multiplex system was validated for sensitivity,specificity,DNA mixtures,precision,reproducibility,stability,anti-inhibition,PCR-based conditions.Results All the validation results showed that the DNATyper17+28Y system would be a sensitive,robust,reliable,stable and reproducible system.Conclusions DNATyper 17+28 Y system can meet the needs of individual identification,kinship analysis,pedigree investigation and so on,which has practical value in forensic.
