摘  要:背景:小凹蛋白1广泛分布于内皮细胞、上皮细胞、血管平滑肌细胞等多种细胞中,在促进内皮细胞增殖及新生血管生成等方面扮演着重要角色,因此其在缺血性疾病中越来越受到重视。目的:基于血管紧张素1-7信号通路探究小凹蛋白1对心肌梗死大鼠心脏微血管、心室重构及线粒体呼吸功能的影响。方法:选取60只雄性SD大鼠,采用随机数字表法分为6组,每组10只:A组为正常对照;B-F组建立心肌梗死模型,造模后12 h,C组尾静脉注射小凹蛋白1空质粒,D组尾静脉注射小凹蛋白1质粒,E组尾静脉注射三氮脒(血管紧张素转换酶2激动剂),F组尾静脉注射小凹蛋白1质粒与三氮脒,1次/d。连续注射28 d后,心脏彩超检测大鼠心功能,心脏凝胶墨汁染色检测微血管密度,苏木精-伊红染色观察大鼠心肌组织形态,氧电极法检测线粒体呼吸功能,免疫印迹法检测小凹蛋白1、血管紧张素1-7信号通路、心脏微血管相关指标的蛋白表达。结果与结论:(1)与A组比较,B组大鼠心功能显著降低(P <0.05);与B组比较,D-F组大鼠心功能均有不同程度改善(P <0.05);F组大鼠心功能好于D、E组(P <0.05);(2)与A组比较,B组大鼠心肌结构遭到破坏,心肌细胞排列杂乱,可见明显炎性细胞浸润,心脏微血管密度显著降低(P <0.05);与B组比较,D-F组大鼠心肌病理形态明显改善,心脏微血管密度升高(P <0.05);F组心脏微血管密度高于D、E组(P <0.05);(3)与A组相比,B组3态呼吸水平及呼吸控制率显著降低(P <0.05),4态呼吸水平显著升高(P <0.05);与B组比较,D-F组3态呼吸水平及呼吸控制率升高(P <0.05),4态呼吸水平降低(P <0.05);与D、E组比较,F组3态呼吸水平及呼吸控制率升高(P <0.05),4态呼吸水平降低(P <0.05);(4)与A组相比,B组小凹蛋白1、血管紧张素转换酶2、血管紧张素1-7、MAS受体、血小板内皮细胞黏附分子1蛋白表达显著降低(P<0.05);与B组比较,D-F组上述蛋白表�BACKGROUND:Caveolin-1 is widely distributed in various kinds of cells,such as endothelial cells,epithelial cells,and vascular smooth muscle cells,and plays an important role in promoting endothelial cell prolife ration and angiogenesis.Therefore,it has been increasingly highlighted in ischemic diseases.OBJECTIVE:To explo re the effects of caveolin-1 on cardiac microvascularization,ventricular remodeling and mitochondrial respiratory function in rats with myocardial infa rction based on angiotensin 1-7 signaling pathway.METHODS:Sixty male Sprague-Dawley rats were randomly divided into normal group(group A),model(group B),empty transfection(group C),Caveolin-1 transfection(group D) and triazamidine(an angiotensin co nverting enzyme 2 agonist;group E),and combination group(Caveolin-1+triazamidine),with 10 rats in each group.Administration in each group was given via the caudal vein,once a day,beginning at 12 hours after modeling.The left co ronary artery ligation method was adopted to establish myocardial infa rction models in the latter five groups.After 28 days of injection,cardiac function of rats was detected by color Doppler ultrasound,microvascular density was detected by cardiac gelink staining,and pathological morphology of myocardial tissue was detected by hematoxylin-eosin staining.Mitochondrial res piratory function was detected by oxygen electrode method.The expression of Caveolin-1,angiotensin 1-7 signaling pathway and cardiac microvascular related indicators were detected by western blot assay.RESULTS AND CONCLUSION:Compared with group A,the cardiac function of rats in group B was significantly reduced(P <0.05).Compared with group B,the cardiac function of rats in groups D-F was improved to different extents(P <0.05).The cardiac function of rats was better in group F than groups D and E(P<0.05).Compared with group A,rats in group B had damaged myocardial structure,irregular arrangement of myocardial cells,markedly inflammato ry cell infiltration,and significantly decreased cardiac microvessel density(P
