摘 要:目的:观察“通督启神”法电针对阿尔茨海默病(AD)模型小鼠空间学习记忆能力和大脑皮层HIF-1α、BNIP3表达的影响,探讨其治疗AD的作用机制。方法:30只6月龄APP/PS1双转基因小鼠随机分为模型组、电针组与电针+抑制剂组,用10只相同遗传背景的C57BL/6小鼠作为正常对照组。采用Morris水迷宫观察各组小鼠行为学变化;免疫组化DAB染色方法观察各组小鼠大脑皮层HIF-1α、BNIP3、Beclin1表达情况;Western blot检测各组小鼠大脑皮层HIF-1α、BNIP3、Beclin1及PI3K的表达含量。结果:在隐蔽平台实验中,电针组逃避潜伏期低于模型组(P<0.05)和电针+抑制剂组,差异无统计学意义(P>0.05),电针+抑制剂组较模型组有下降趋势,差异无统计学意义(P>0.05);在空间探索实验中,电针组目标象限进入次数和游泳距离明显大于模型组和电针+抑制剂组,电针+抑制剂组较模型组有上升趋势,差异无统计学意义(P>0.05);免疫组化DAB染色结果表明,电针组HIF-1α、BNIP3表达高于模型组和电针+抑制剂组,差异具有统计学意义(P<0.05),电针+抑制剂组较模型组有上升趋势,差异无统计学意义(P>0.05);Western Blot检测结果表明,电针组HIF-1α、BNIP3表达均高于模型组和电针+抑制剂组,差异具有统计学意义(P<0.05),电针+抑制剂组较模型组相比有上升趋势,差异无统计学意义(P>0.05)。结论:“通督启神”法电针能够提高APP/PS1小鼠的空间学习记忆能力,并增加大脑皮层HIF-1α、BNIP3的表达,可能与增加线粒体自噬机制相关。Objective:To observe the effects of electro-acupuncture(EA)on spatial learning and memory ability and expressions of HIF-1αand BNIP3 in cerebral cortex of the mice with Alzheimer’s disease(AD)based on‘Tongdu Qishen’theory,and to explore its mechanism in treating AD.Methods:30 APP/PS1 double transgenic mice at 6-month-old were randomly divided into the model group,the EA group,the EA plus inhibitor group,and 10 C57BL/6 mice with the same genetic background were taken as the normal control.Morris water maze test was used to observe the ethology changes of mice in each group.Immunohistochemical DAB staining was used to observe the expressions of HIF-1α,BNIP3 and Beclin1 in cerebral cortex of mice in each group.Western blot was used to observe the expressions of HIF-1α,BNIP3,Beclin1 and PI3K in cerebral cortex of mice in each group.Results:In concealed platform experiment,the escape latency in the EA group was lower than that in the model group(P<0.05),and the escape latency in the EA plus inhibitor group showed descending tendency compared with that in the model group(P>0.05).In the space exploration experiment,the entry times and swimming distance of the target quadrant in the EA group were significantly more than those in the model group and the EA plus inhibitor group,and the EA plus inhibitor group had an upward trend compared with the model group(P>0.05).Immunohistochemical DAB staining results showed that the expressions of HIF-1αand BNIP3 in the EA group were higher than those in the model group and the EA plus inhibitor group(P<0.05),and the expressions of HIF-1αand BNIP3 in the EA plus inhibitor group showed ascending trend compared with those in the model group(P>0.05).Western blot showed that the expressions of HIF-1αand BNIP3 in the EA group were higher than those in the model group and the EA plus inhibitor group(P<0.05),and the expression of HIF-1αand BNIP3 in the EA plus inhibitor group showed ascending trend compared with those in the model group(P>0.05).Conclusion:‘Tongdu Qishen’EA
