摘　　要：目的探讨克唑替尼致心动过缓的临床特点及可能机制,为临床安全用药提供参考。方法检索国内外文献数据库,收集克唑替尼致心动过缓的文献报道,检索时限为建库至2022年5月,提取文献中患者的基本信息、用药情况、药品不良反应(ADR)发生过程和转归等信息进行分析。结果共检索到10例患者,男5例,女5例,平均年龄(60.9±17.5)岁。7例ADR在用药后4个月内发生,10例患者用药后心率均降至60次/min以下,其中6例患者心率降至45次/min以下,最低降至24次/min。10例患者的心动过缓均与克唑替尼相关,其中7例患者为严重的ADR,停药或对症治疗后好转。结论克唑替尼致心动过缓虽为常见的不良反应,但目前文献报道较少,且多为严重ADR,需引起临床关注与重视。Objective To investigate the clinical characteristics and possible mechanisms of bradycardia related to crizotinib,and to provide a reference for safe clinical medication.Methods Literatures on bradycardia induced by crizotinib were searched from domestic and foreign literature databases until May 2022.The data from the literature were collected and analyzed,including patients′basic data,medication data,development of adverse drug reactions and outcomes.Results A total of 10 cases were screened out.Among them,5 cases were male and 5 were female,and the average age was(60.9±17.5)years old.Seven patients had ADR within 4 months after taking crizotinib.The heart rate(HR)of all 10 patients dropped below 60,among which the HR of 6 patients dropped below 45,and the lowest rate was 24.All patients suffered from bradycardia caused by crizotinib and 7 patients developed serious ADR,and they all recovered after treatment.Conclusion Although crizotinib-induced bradycardia is common ADR,there are few literature reports,and most of them are serious ADR.Great importance should be attached to the ADR in clinic.
