巴豆生物碱调控miR-4317对结直肠癌SW620细胞增殖和凋亡的影响及机制

摘　　要：目的探讨巴豆生物碱(crotonic alkaloids,CA)对结直肠癌SW620细胞增殖和凋亡的影响及可能的作用机制。方法体外培养人结直肠癌细胞SW620,不同浓度的CA处理SW620细胞,脂质体转染法将miR-NC、miR-4317 mimics分别转染至SW620细胞,将anti-miR-NC、anti-miR-4317分别转染至SW620细胞后加入100μg/ml巴豆生物碱处理24 h;MTT实验检测细胞增殖;平板克隆形成实验检测细胞克隆形成能力;流式细胞术检测细胞凋亡率;qRT-PCR法检测miR-4317的表达量。结果CA处理SW620细胞后,miR-4317的表达量和细胞增殖抑制率升高(P<0.05),细胞凋亡率升高(P<0.05),细胞克隆形成数减少(P<0.05),且呈剂量依赖性;转染miR-4317 mimics后,细胞增殖抑制率升高(P<0.05),细胞凋亡率升高(P<0.05),细胞克隆形成数减少(P<0.05);转染anti-miR-4317可降低CA对SW620细胞增殖及凋亡的作用。结论巴豆生物碱可通过上调miR-4317的表达而抑制结直肠癌细胞增殖并可诱导细胞凋亡。Objective To investigate the effects of croton alkaloids(CA)on the proliferation and apoptosis of colorectal cancer SW620 cells by regulating the expression of miR-4317 in vitro,and to explore its action mechanism.Methods Human colorectal cancer cells SW620 were cultured in vitro,and SW620 cells were treated by different concentrations of CA.The liposome transfection method was used to transfect miR-NC and miR-4317 mimics into SW620 cells,respectively,and anti-miR-NC and anti-miR-4317 were respectively transfected into SW620 cells and treated by 100μg/ml CA for 24h.MTT was used to detect cell proliferation;plate clone formation test was used to detect the ability of cell clone formation;flow cytometry was used to detect the apoptosis rate,and qRT-PCR was used to detect the expression levels of miR-4317.Results After SW620 cells were treated by CA,the expression levels of miR-4317 and cell proliferation inhibition rate as well as apoptosis rate were significantly increased(P<0.05),however,the numbers of cell cloneformation were significantly decreased(P<0.05),with a dose-dependent manner.After transfection of miR-4317 mimics,the cell proliferation inhibition rate and cell apoptosis rate were significantly increased(P<0.05),burt,the numbers of cell clone formation w were significantly decreased(P<0.05).Transfection of anti-miR-4317 could decrease the effects of CA on the proliferation and apoptosis of SW620 cells.Conclusion The croton alkaloids can inhibit the proliferation of colorectal cancer cells and induce apoptosis by up-regulating the expression of miR-4317.
