涤痰解毒方对急性有机磷农药中毒大鼠阿托品化后血清炎症因子水平和重要脏器超微结构的影响

摘　　要：目的探讨涤痰解毒方对急性有机磷农药中毒(AOPP)大鼠阿托品化后血清炎症因子水平和重要脏器超微结构的影响。方法将18只大鼠随机分为正常组、模型组、西药组、等剂量涤痰解毒方组、高剂量涤痰解毒方组、西药+等剂量涤痰解毒方组,每组3只。除正常组外,给予其余组灌胃敌敌畏溶液建立AOPP模型,建模成功后给予肌内注射阿托品+解磷定直至阿托品化。阿托品化后,西药组给予肌内注射长托宁,等剂量涤痰解毒方组、高剂量涤痰解毒方组分别按14 g/kg、28 g/kg给予大鼠灌胃涤痰解毒方,西药+等剂量涤痰解毒方组大鼠给予肌内注射长托宁和灌胃涤痰解毒方(14 g/kg),模型组给予灌胃等体积温开水。治疗24 h后检测各组大鼠血清IL-1β、IL-6水平,并取大脑皮层组织、海马组织及肺组织在电镜下观察其超微结构。结果模型组血清IL-1β水平高于正常组,西药+等剂量涤痰解毒方组、等剂量涤痰解毒方组、高剂量涤痰解毒方组的血清IL-1β水平低于模型组(均P<0.05),但各药物干预组间差异无统计学意义(均P>0.05)。6组间血清IL-6水平差异无统计学意义(P>0.05)。模型组大鼠肺组织、海马组织、大脑皮质组织结构损坏明显,各药物干预组大鼠上述组织结构的破坏均有一定程度的改善。结论涤痰解毒方可能通过减轻炎症反应,改善肺脏、海马、大脑皮质等重要脏器结构,促进AOPP大鼠阿托品化后机体的恢复。Objective To explore the effect of Ditan Jiedu Prescription on serum inflammatory factor levels and vital organs ultrastructure in rats with acute organophosphorus pesticide poisoning(AOPP)after atropinization.Methods A total of 18 rats were randomly assigned to normal group,model group,Western Medicine group,equivalent-dose Ditan Jiedu Prescription group,high-dose Ditan Jiedu Prescription group,or Western Medicine+equivalent-dose Ditan Jiedu Prescription group,with 3 rats in each group.Except for the normal group,the remaining groups received intragastric administration of dichlorvos solution to construct the AOPP model,and intramuscular injection of atropine+pralidoxime iodide was given to the rats until atropinization after successful modeling.After atropinization,the intramuscular injection of penehyclidine hydrochloride was given to the Western Medicine group.The equivalent-dose,high-dose Ditan Jiedu Prescription groups received intragastric administration of 14 g/kg,28 g/kg Ditan Jiedu Prescriptions,respectively.The Western Medicine+equivalent-dose Ditan Jiedu Prescription groups received intramuscular injection of penehyclidine hydrochloride and intragastric administration of Ditan Jiedu Prescription(14 g/kg).The model group received intragastric administration of equivalent volume of warm water.The levels of serum interleukin(IL)-1β,IL-6 of rats were detected after 24 hours of treatment.Rats′cerebral cortex tissues,hippocampus and pulmonary tissues were removed for the observation of their ultrastructure under the electron microscope.Results The level of serum IL-1βin the model group was higher than that in the normal group,and the level of serum IL-1βin the Western Medicine+equivalent-dose Ditan Jiedu Prescription group,equivalent-dose Ditan Jiedu Prescription group,and high-dose Ditan Jiedu Prescription group was lower than that in the model group(all P<0.05);however,no statistically significant difference in drugs intervention was found between the various groups(all P>0.05).There was no statistic
