点篮子鱼形态特征及6种组织乳酸脱氢酶的电泳分析

摘　　要：为了解点篮子鱼(Siganus guttatus)的形态特征和生化遗传特性,为其种质鉴定和制定种质标准提供理论依据。采用传统形态学方法测定样品的可量性状和可数性状,通过聚丙烯酰胺凝胶垂直板电泳对30尾点篮子鱼的6种组织(心脏、眼睛晶状体、肌肉、肝脏、肾脏和脾脏)的乳酸脱氢酶(LDH)进行分析。结果表明:1)点篮子鱼的主要形态特征为体呈椭圆形,侧扁,背高,体背缘和腹缘浅弧形,头较小,口下位;鳍式为背鳍D.Ⅻ~XIV-9~11,胸鳍P.ⅱ-14~15,腹鳍V.Ⅰ-3-Ⅰ,臀鳍A.Ⅶ~Ⅷ-8~10;左侧第一鳃弓外侧鳃耙数为23~26。身体两侧上半部褐色,分布着许多橙黄色斑块,下半部灰褐色,背鳍、胸鳍和腹鳍浅黄色,臀鳍和尾鳍浅灰褐色。2)点篮子鱼的心脏、眼睛晶状体、肌肉、肝脏、肾脏和脾脏中的LDH酶带条数分别为:4、7、2~3、1~3、3~4和1~3条。点篮子鱼的乳酸脱氢酶具有组织特异性,眼睛晶状体LDH可作为鉴定点篮子鱼种质的特征生化遗传参数。This paper is to study the morphological characteristics and biochemical genetic characteristics of Siganus guttatus so as to provide a theoretical basis for their germplasm identification and the establishment of the germplasm standards.The countable and measurable characters of Siganus guttatus were measured by the traditional morphological methods.By using the polyacrylamide gel electrophoresis,the lactate dehydrogenase(LDH)isozymes expressed in six tissues(heart,eye,muscle,liver,kidney and spleen)of 30 fish samples.were analysed.The results showed several main morphological characteristics as follows.⑴The body of Siganus guttatus was oval,flat,and fusiform,with high dorsum,smaller head and lower mouth.The dorsal and ventral side had a shallow arc shape.The fin type was dorsal fin D.Ⅻ~XIV-9~11,pectoral fin P.ⅱ-14~15,ventral fin V.Ⅰ-3-Ⅰand anal fin A.Ⅶ~Ⅷ-8~10.The number of gill rakers on the left side of the first lateral arch is 23~26.The upper parts of the body were tan with lots of orange spots,and the lower parts of the body were grey and tan.The dorsal fin,pectoral fin and ventral fin were pale-yellow,and the anal fin and caudal fin pale were grey and tan.⑵The electrophoretic band numbers of lactate dehydrogenase(LDH)isozymes expressed in heart,eye,muscle,liver,kidney and spleen were 4,7,2~3,1~3,3~4 and 1~3,respectively.The LDH isozymes of Siganus guttatus presented tissue specificity.And the LDH isozymes expressed in eyes can be used as the biochemical genetic marker for identifying Siganus guttatus.
