摘 要:本研究建立了水产苗种中喹乙醇(Olaquindox,OLA)代谢物3-甲基-喹噁啉-2-羧酸(3-methyl-quinoxaline-2-carboxylic acid,MQCA)残留量检测的酶联免疫法(ELISA)。乙酸乙酯作为提取溶剂,采用盐酸(HCl)溶液对样品提取液酸解、纯水饱和正己烷脱脂除杂等措施对前处理过程进行优化。结果表明,MQCA在0.400~60.000μg/kg范围内线性良好,相关系数R^(2)=0.996,检出限为0.400μg/kg。在4.00、8.00、40.00μg/kg的加标水平下,中国对虾(Penaeus chinensis)、三疣梭子蟹(Portunus trituberculatus)、刺参(Stichopus japonicus)、牙鲆(Paralichthys olivaceus)等不同基质苗种中MQCA的平均回收率为71.4%~95.7%,精密度为3.9%~7.9%。与HPLC法相比,该法灵敏度高、特异性强、快速简便,适用于不同基质水产苗种中MQCA残留量的快速批量检测。An enzyme-linked immunoassay(ELISA)was established for the determination of 3-methyl-quinoxaline-2-carboxylic acid(MQCA)residues in aquatic seedlings.Ethyl acetate was used as the extraction solvent,HCl solution was used to acid hydrolyze the sample extract,and pure water saturated n-hexane was used to degrease and remove impurities,which optimized the sample pretreatment process.The results showed that MQCA had good linear in the range of 0.400~60.000μg/kg,with the correlation coefficients R ^(2)=0.996,and the detection limit was 0.400μg/kg.At the spiked level of 4.00,8.00 and 40.00μg/kg,the average recovery of MQCA in different substrates including Penaeus chinensis,Portunus trituberculatus,Stichopus japonicus and Paralichthys olivaceus in was 71.4%~95.7%,and the precision was 3.9%~7.9%.Compared with HPLC,this method had the advantages of high sensitivity,strong specificity,rapidity and simplicity,and was suitable for rapid batch determination of MQCA residues in aquatic seedlings in different substrates.
