摘 要:目的:研究β-榄香烯抑制结直肠癌细胞侵袭和转移的作用及可能的机制。方法:用20、40、80、160、320μg/mL的β-榄香烯处理人结肠癌HCT116细胞,采用MTT法检测细胞的增殖抑制率,划痕实验检测细胞迁移率,Transwell小室体外迁移实验检测细胞迁移率,Transwell小室体外侵袭实验检测细胞侵袭率;蛋白免疫印记法(Western blotting)检测细胞Vimentin、E-cadherin、N-cadherin、β-catenin表达;实时荧光定量聚合酶链式反应(Realtime PCR)检测细胞Vimentin mRNA、E-cadherin mRNA、N-cadherin mRNA、β-catenin mRNA表达。结果:β-榄香烯浓度越高,处理时间越长,抑制HCT116细胞增殖活性越明显(P<0.05)。与空白组相比,80μg/mLβ-榄香烯处理后HCT116细胞迁移率、侵袭率、划痕愈合率,N-cadherin、Vimentin、β-catenin蛋白表达降低,E-cadherin蛋白表达升高,细胞增殖抑制率升高(P<0.05)。结论:β-榄香烯可能通过抑制人结肠癌细胞系HCT116中EMT信号通路来抑制细胞增殖,并抑制其侵袭转移能力。Objective:To study the inhibitory effect ofβ-elemene on invasion and metastasis of colorectal cancer cells and its possible mechanism.Methods:Human colon cancer HCT116 cells were treated with 20,40,80,160 and 320μg/mLβ-elemene.Proliferation inhibition rate of the cells was detected by MTT assay,cell migration rate was detected by scratched assay.Transwell cell migration test was used to detect the cell migration rate,Transwell cell invasion test was used to detect the cell invasion rate.The expression of Vimentin,E-cadherin,N-cadherin andβ-catenin were detected by Western blotting.The mRNA expressions of Vimentin,e-cadherin,N-cadherin andβ-catenin were detected by Real-time PCR.Results:The higher the concentration ofβ-elemene,the longer the treatment time,the more obvious the inhibition of HCT116 cell proliferation activity(P<0.05).Compared with blank control group,the expression of migration rate,invasion rate,scratch healing rate,N-cadherin,Vimentin andβ-catenin protein of HCT116 cells were decreased after treated with 80μg/mLβ-elemene,the expression of E-cadherin protein was increased,and the inhibition rate of cell proliferation was increased(P<0.05).Conclusion:β-elemene may inhibit cell proliferation and invasion and metastasis by inhibiting EMT signaling pathway in human colon cancer cell line HCT116.
