摘 要:目的 探究集落刺激因子1(colony stimulating factor 1, CSF1)/集落刺激因子1受体(colony stimulating factor 1receptor, CSF1R)信号在控制夏氏疟原虫(Plasmodium chabaudi, P. chabaudi)再燃中的作用,并对其作用机制进行初步探讨。方法ELISA检测P. chabaudi感染不同时间小鼠血清CSF1浓度变化;随后采用CSF1及CSF1R体内抗体中和实验,观察CSF1/CSF1R信号在控制夏氏疟原虫再燃中的作用;最后通过流式细胞术观察CSF1/CSF1R信号作用的可能效应细胞。结果 与未感染小鼠相比,P. chabaudi感染小鼠血清CSF1浓度随感染时间的延长而逐渐升高,并在再燃阶段达到最高水平。在体分别中和CSF1或CSF1R后,感染小鼠再燃阶段的原虫血症显著增高;虽然固有免疫细胞表面CSF1R的表达不随感染升高,但中和CSF1和CSF1R后能显著降低感染小鼠脾脏非经典单核细胞(non classical monocytes, NCMs)和红髓巨噬细胞(red pulp macrophages,RPMs)频率(P<0.05)。结论 CSF1可能通过作用于NCMs和RPMs细胞表面的CSF1R来维持其细胞数目,从而在控制夏氏疟原虫再燃中发挥重要作用。This study was performed to explore the effect of CSF1/CSF1R signaling on the recurrence stage of Plasmodium chabaudi infection and its mechanism. ELISA showed demonstrated that the CSF1 concentration in the serum of mice infected with P. chabaudi increased gradually with the prolongation of infection time, and reached the highest level at the recurrence stage. The parasitemia at the recurrence stage were significantly increased, after the CSF1 or CSF1R was neutralized. Although CSF1R expressed on the surface of innate immune cells would not increase with infection, the neutralization of CSF1 and CSF1R significantly reduced the frequency of both nonclassical monocytes(NCMs) and red pulp macrophages(RPMs) in the spleen of infected mice(P<0.05). Therefore,CSF1/CSF1R signaling might play an important role in the control of P. chabaudi recurrence by maintaining the number of NCMs and RPMs.
