摘 要:目的探讨salusinβ能否减轻内皮素-1(ET-1)诱导的离体心脏功能损伤。方法按照随机对照表法将SD大鼠分为对照组、ET-1组和salusinβ+ET-1组,每组各6只;采用Langendorff系统构建ET-1诱导离体大鼠心脏灌流损伤模型,比较各组心率(HR)、左室舒张末压(LVEDP)、左室收缩压(LVSP)、左心室内压最大上升/下降速率(±dp/dtmax)、冠状动脉流量(CF)。结果灌流过程中3组HR和LVEDP差异无统计学意义(均P>0.05);与对照组比较,ET-1组ET-1灌流后LVSP、+dp/dt、-dp/dt急剧下降,灌流10 min后LVSP、+dp/dt、-dp/dt分别为(61.65±6.17)mmHg、(1138±48)mmHg·s^(-1)和(818±57)mmHg·s^(-1);Salusinβ+ET-1组给予ET-1灌流10 min后LVSP、+dp/dt、-dp/dt分别为(90.89±7.44)mmHg、(1573±89)mmHg·s^(-1)和(1178±78)mmHg·s^(-1),均显著高于ET-1组(均P<0.05)。与对照组比较,ET-1组灌流后CF急剧下降,ET-1灌流后5、10、15 min CF分别为(7.1±0.5)mL、(5.9±0.4)mL和(5.8±0.4)mL,Krebs-Henseleit(KH)液灌流后CF有所上升,但仍明显低于对照组(均P<0.01);Salusinβ+ET-1组灌流后CF均下降,但ET-1灌流后5、10、15 min CF分别为(10.1±0.3)mL、(9.1±0.4)mL、(9.4±0.5)mL,均显著高于ET-1组(均P<0.01)。结论Salusinβ可减轻ET-1诱导的离体心脏功能损伤。Objective To explore whether salusinβcan alleviate endothelin-1(ET-1)-induced cardiac function injury in vitro.Methods Eighteen SD rats were randomly divided into control group,ET-1 group and salusinβ+ET-1 group,with 6 rats in each group.The isolated rat hearts were perfused with Langendorff system to establish the perfusion injury model induced by ET-1.The heart rate(HR),left ventricular end diastolic pressure(LVEDP),left ventricular systolic pressure(LVSP),coronary flow(CF),and maximum rate of increase/decrease of left ventricular pressure(±dp/dtmax)were recorded in all groups.Results There were no significant differences in HR and LVEDP among the three groups during the perfusion(P>0.05).Compared with the control group,ET-1 perfusion induced a sharp decrease in LVSP,+dp/dt and-dp/dt,which were(61.65±6.17)mmHg,(1138±48)mmHg·s^(-1) and(818±57)mmHg·s^(-1) at 10 min after perfusion,respectively.In salusinβ+ET-1 group,the LVSP,+dp/dt and-dp/dt were,respectively,(90.89±7.44)mmHg,(1573±89)mmHg·s^(-1) and(1178±78)mmHg·s^(-1),which were significantly higher than those in ET-1 group(P<0.05).Compared with the control group,the CF decreased sharply in ET-1 group.The CF was(7.1±0.5)mL,(5.9±0.4)mL and(5.8±0.4)mL at 5,10 and 15 min after ET-1 perfusion,respectively.Although the CF in ET-1 group increased after KH solution perfusion,it was still lower than that in the control group(P<0.01).In salusinβ+ET-1 group,the CF was(10.1±0.3)mL,(9.1±0.4)mL and(9.4±0.5)mL at 5,10 and 15 min after ET-1 perfusion,respectively,which were significantly higher than those in the ET-1 group(P<0.01).Conclusion Salusinβcan alleviate ET-1-induced cardiac function injury in vitro.
