摘 要:目的:研究靶向超声微泡介导血管内皮生长因子(VEGF)基因转染对薄型子宫内膜容受性(ER)的影响。方法:选择40只性成熟但未交配的具有动情周期的雌性SD大鼠进行研究。将其随机分为四组,包括正常组10只,模型组10只,阴性对照组10只,治疗组10只。其中模型组、阴性对照组及治疗组大鼠均自宫角处注入95%无水乙醇贮留5 min制备薄型子宫模型,正常组自宫角处注入生理盐水,贮留5 min。模型组予以生理盐水注射治疗,阴性对照组予以超声微泡+抗体(不含基因)治疗,治疗组则以靶向超声微泡介导VEGF基因转染治疗。比较四组大鼠子宫内膜厚度及腺体数量,子宫内膜血流灌注,子宫内膜VEGF及白血病抑制因子(LIF)水平。结果:正常组子宫内膜厚度及腺体数量均高于模型组、阴性对照组及治疗组,治疗组子宫内膜厚度及腺体数量均高于模型组、阴性对照组(P<0.05)。正常组阻力指数(RI)、搏动指数(PI)、脐动脉收缩压与舒张压比值(S/D)均低于模型组、阴性对照组及治疗组,治疗组RI、PI、S/D均低于模型组、阴性对照组(P<0.05)。正常组子宫内膜VEGF、LIF水平均高于模型组、阴性对照组及治疗组,治疗组子宫内膜VEGF、LIF水平均高于模型组、阴性对照组(P<0.05)。结论:靶向超声微泡介导VEGF基因转染可有效修复子宫内膜,改善ER,值得临床重点关注。Objective:To investigate the effect of targeted ultrasound microbubble-mediated vascular endothelial growth factor(VEGF)gene transfection on the thin endometrial receptivity(ER).Method:A total of 40 sexually mature but unmated female SD rats with estrous cycles were selected for this study.They were randomly divided into four groups,including 10 rats in the normal group,10 rats in the model group,10 rats in the negative control group,and 10 rats in the treatment group.The rats in the model group,negative control group and treatment group were all injected with 95%anhydrous ethanol from the uterine horn and stored for 5 min to prepare the thin uterine model.The rats in the normal group were injected with normal saline from the uterine horn and stored for 5 min.The model group was treated with normal saline injection,the negative control group was treated with ultrasound microbubble+antibody(without gene),and the treatment group was treated with targeted ultrasound microbubblemediated VEGF gene transfection.The endometrial thickness and gland number,endometrial bloodstream perfusion,and endometrial VEGF and leukaemia inhibitory factor(LIF)levels in the four groups were compared.Result:The endometrial thickness and gland number in the normal group were higher than those in the model group,negative control group,and treatment group,the endometrial thickness and gland number in the treatment group were higher than those in the model group and negative control group(P<0.05).The resistance index(RI),pulsatility index(PI)and the ratio of umbilical artery systolic to diastolic blood pressure(S/D)values in the normal group were lower than those in the model group,negative control group,and treatment group,the RI,PI,and S/D values in the treatment group were lower than those in the model group and negative control group(P<0.05).The levels of VEGF and LIF in endometrium of the normal group were higher than those in the model group,negative control group and treatment group,and the levels of VEGF and LIF in endometrium of the
