摘 要:目的探讨滋肾通关方中肉桂反佐配伍对黄柏-知母药对的活性成分在正常大鼠体内组织分布的影响及其作用机制。方法将56只Wistar大鼠随机分为空白组(等量生理盐水),滋肾通关方低、中、高剂量组(5,10,20 g/kg),黄柏-知母药对低、中、高剂量组(5,10,20 g/kg),各8只。分别灌胃相应药物1 h后取大鼠心、肝、脾、肺、肾组织并制备匀浆液,采用超高效液相色谱串联质谱法测定大鼠体内组织匀浆液中黄柏碱和知母皂苷AⅢ的含量。结果与黄柏-知母药对低、中、高剂量组比较,滋肾通关方低、中、高剂量组大鼠心、肝、脾、肺、肾组织匀浆液中黄柏碱和知母皂苷AⅢ的含量整体更高;滋肾通关方高剂量组大鼠肺组织匀浆液中黄柏碱和知母皂苷AⅢ的含量显著高于黄柏-知母药对高剂量组(P<0.05),滋肾通关方低剂量组大鼠心组织中黄柏碱的含量显著高于黄柏-知母药对低剂量组(P<0.05)。结论滋肾通关方中肉桂反佐配伍黄柏-知母药对的作用机制可能与改变药对中活性成分在组织中的分布有关。Objective To investigate the effect of corrigent compatibility of Cinnamomi Cortex in Zishen Tongguan Recipe on the tissue distribution of active ingredients of Phellodendri Chinensis Cortex-Anemarrhenae Rhizoma herb pair in normal rats,and to study its mechanism.Methods Fifty-six Wistar rats were selected and randomly divided into the blank group(equal amount of normal saline),Zishen Tongguan Recipe low-,medium-and high-dose groups(5,10,20 g/kg),Phellodendri Chinensis Cortex-Anemarrhenae Rhizoma herb pair low-,medium-and high-dose groups(5,10,20 g/kg),with eight rats in each group.All rats were given corresponding drugs by gavage respectively.After 1 h of administration,the heart,liver,spleen,lung and kidney tissues of rats were taken and homogenized,and the contents of phellodendrine and timosaponin AⅢin the tissue homogenates were determined by the ultra-high-performance liquid chromatography-tandem mass spectrometry(UHPLC-MS/MS)method.Results Compared with those in the Phellodendri Chinensis Cortex-Anemarrhenae Rhizoma herb pair low-,medium-and high-dose groups,the contents of phellodendrine and timosaponin AⅢin the homogenates of heart,liver,spleen,lung and kidney tissues in the Zishen Tongguan Recipe low-,medium-and high-dose groups were higher.The contents of phellodendrine and timosaponin AⅢin the lung homogenate in the Zishen Tongguan Recipe high-dose group were significantly higher than those in the Phellodendri Chinensis Cortex-Anemarrhenae Rhizoma herb pair high-dose group(P<0.05),and the content of phellodendrine in heart tissue in the Zishen Tongguan Recipe low-dose group was significantly higher than that in the Phellodendri Chinensis Cortex-Anemarrhenae Rhizoma herb pair low-dose group(P<0.05).Conclusion The mechanism of corrigent compatibility of Cinnamomi Cortex with Phellodendri Chinensis Cortex-Anemarrhenae Rhizoma herb pair in Zishen Tongguan Recipe may be related to the change of the tissue distribution of active ingredients of the herb pair.
