摘 要:目的观察大鼠围产期低蛋白饲料对子代的体质量、葡萄糖代谢及肠道葡萄糖转运体的影响。方法将12只妊娠期大鼠随机分为标准组和低蛋白组,每组6只,每只大鼠独立笼饲养,在3周的妊娠期和3周的哺乳期分别给予标准饲料或低蛋白饲料。在子鼠出生后,每笼留4只雄鼠,子鼠3周龄离乳后均给予标准饲料。每周测量子鼠体质量。将分娩日设为子鼠出生第0天,在第3周和第10周,每笼中随机选取1只子鼠行葡萄糖耐量试验;另1只子鼠测定血清中糖脂代谢相关激素:胰岛素、胰高糖素样肽(GLP-1)和酪氨酸-酪氨酸多肽(PYY)浓度,并收集其空肠、回肠及十二指肠组织,以检测肠道葡萄糖转运基因:钠依赖葡萄糖转运蛋白1(SGLT1)、葡萄糖转运蛋白2(GLUT2)和葡萄糖转运蛋白5(GLUT5)的表达。结果与标准组比较,低蛋白组子鼠出生时体质量降低,并持续到10周龄(P<0.05)。与标准组比较,低蛋白组子鼠在10周龄时糖耐量减退(P<0.05)。在3周龄时,低蛋白组子鼠血清中GLP-1的浓度较标准组降低(P<0.05);在10周龄时,低蛋白组血清中胰岛素、GLP-1、PYY的浓度均较标准组降低(P<0.05)。在3周龄和10周龄,低蛋白组子鼠小肠组织中SGLT1、GLUT2和GLUT5的表达水平较标准组增加(P<0.05)。结论母代在围产期的低蛋白饮食会增加子代肠道葡萄糖转运体的表达,促进肠道对葡萄糖吸收,加重胰岛细胞负担,导致子代的糖耐量减退,即使子代离乳后给予标准饮食也不能完全逆转。Objective To investigate the effects of maternal protein-restricted(PR)diet on offspring’s body weight,glucose metabolism,and intestinal glucose transporter.Methods Pregnant Sprague-Dawley rats were divided randomly into standard diet group and PR diet group,with 6 rats in each group.Rats were raised individually in conventional cages and fed with standard chow diet and PR diet during 3-week pregnancy and 3-week lactation,respectively.Four male offspring were left in each cage after birth.The offspring were fed with standard diet after being weaned at 3 weeks of age.The day of parturition was set as postnatal day 0.The body weight of offspring was measured weekly.At 3 weeks and 10 weeks of age,an offspring was randomly selected from each cage to perform the oral glucose tolerance tests;and another offspring was randomly selected from each cage to determine the serum concentrations of glucose and lipid metabolism-related hormones[insulin,glucagon-like peptide(GLP-1)and tyrosine-tyrosine polypeptide(PYY)],and then duodenum,jejunum and ileum were collected to detect the gene expression of intestinal glucose transporter[sodium-dependent glucose transporter 1(SGLT1),glucose transporter 2(GLUT2)and GLUT5].Results Compared with standard chow diet group,the body weight was significantly decreased at postnatal day 0 in PR diet group,and persisted till 10 weeks of age(P<0.05).Compared with standard chow diet group,the glucose tolerance of PR offspring was decreased at 10 weeks of age in PR diet group(P<0.05).The concentration of plasma GLP-1 at 3 weeks of age was lower in PR group than in standard diet group(P<0.05),the concentrations of plasma insulin,GLP-1 and PYY at 10 weeks of age were lower in PR group than in standard group(P<0.05).Compared with standard group,the mRNA expression levels of SGLT1,GLUT2 and GLUT5 were increased in the intestine tissues of PR offspring at 3 weeks of age and 10 weeks of age in PR diet group(P<0.05).Conclusion These results suggest that maternal PR diet during pregnancy and lactation ma
