摘 要:目的研究Wee1在小鼠1-细胞期受精卵发育全过程(G_(1)、S、G_(2)、M期)中的表达和定位,为进一步探究Wee1对小鼠胚胎有丝分裂(主要是G_(2)/M转换)的作用提供理论依据。方法应用qRT-PCR和免疫印迹技术分别检测小鼠1-细胞期受精卵在G_(1)、S、G_(2)、M不同时期Wee1的表达谱,并利用间接免疫荧光观察Wee1蛋白在1-细胞期受精卵发育全过程中的定位。结果Wee1 mRNA和蛋白在不同时期的表达水平变化趋势一致:从G_(1)到G_(2)期表达水平持续升高(P<0.05),但进入M期后表达水平显著降低(P<0.01)。间接免疫荧光显示:Wee1蛋白在G_(1)、S期定位于细胞质,在G_(2)期Wee1蛋白开始向核转位,M期细胞核内Wee1蛋白明显增多。结论小鼠1-细胞期受精卵的发育伴随Wee1表达水平的改变和Wee1蛋白的核浆转位,提示Wee1表达水平及其蛋白亚细胞定位的改变可能是调节小鼠1-细胞期受精卵细胞进程的机制之一。Objective To investigate the expression and location of Wee1 during the development of mouse one-cell stage zygote(G_(1),S,G_(2),M),so as to provide a theoretical basis for the further study on the role of Wee1 in embryonic mitosis(mainly G_(2)/M transition)in mice.Methods The expression profiles of Wee1 in mouse one-cell stage zygote at G_(1),S,G_(2) and M stages were detected using qRT-PCR and Western blot,respectively.The location of Wee1 protein during the development of one-cell stage zygote was observed by indirect immunofluorescence.Results The mRNA and protein expression levels of Wee1 at different stages showed the same trend:the expression continued to increase from G_(1) to G_(2) stage(P<0.05),but significantly decreased after entering M stage(P<0.01).Indirect immunofluorescence revealed that Wee1 protein was localized in the cytoplasm at G_(1) and S stages,began to translocate to the nucleus at G_(2) stage,and significantly increased in the nucleus at M stage.Conclusion The development of mouse one-cell stage zygote is accompanied by the changes of Wee1 expression and the nucleocytoplasmic translocation of Wee1 protein,suggesting that the changes of Wee1 expression and its subcellular location may be one of the mechanisms for regulating the development of mouse one-cell stage zygote.
