摘 要:目的分析老年2型糖尿病合并轻度认知功能障碍与载脂蛋白E(apolipoprotein E,APOE)、溶质载体有机阴离子转运蛋白家族成员1B1(solute carrier organic anion transporter family member 1B1,SLCO1B1)基因多态性的相关性。方法纳入老年2型糖尿病合并轻度认知功能障碍(病例组)45例和老年2型糖尿病认知功能正常(对照组)34例,比较两组间的APOE、SLCO1B1基因多态性分布。采用χ^(2)检验分析基因型和等位基因在病例组与对照组的分布情况,采用Logistic回归分析老年2型糖尿病患者合并轻度认知功能障碍的影响因素。结果病例组和对照组APOE等位基因和基因型分布差异均有统计学意义(P<0.05)。两组间SLCO1B1等位基因频率分布差异无统计学意义(P>0.05),两组间SLCO1B1的388A>G位点基因型分布差异无统计学意义(P>0.05),521T>C位点基因型分布差异有统计学意义(P<0.05)。Logistic回归分析显示,SLCO1B1基因型与老年T2DM合并MCI无明显相关性(P>0.05),APOE等位基因和APOE基因型对老年T2DM合并MCI的发病有影响,差异有统计学意义(P<0.05)。结论APOE基因多态性与老年2型糖尿病合并轻度认知功能障碍有相关性,APOE等位基因和基因型是老年2型糖尿病合并轻度认知功能障碍的独立危险因素,SLCO1B1基因多态性与老年2型糖尿病合并轻度认知功能障碍无相关性。Objective To analyze the relationship between mild cognitive impairment and gene polymorphism of apolipoprotein E(APOE),solute carrier organic anion transporter family member 1B1(SLCO1B1)in elderly type 2 diabetes patients.Methods Forty five elderly type 2 diabetes patients with mild cognitive impairment(case group)and 34 elderly type 2 diabetes patients with normal cognitive function(control group)were included.Theχ^(2) test was used to analyze the distribution of genotype frequencies and allele frequencies between cases and controls,and Logistic regression was used to analyze the influencing factors of mild cognitive impairment in elderly patients with type 2 diabetes.Results The distribution of APOE gene alleles and genotypes showed no statistical difference between case group and control group(P<0.05).There was no statistical difference in the distribution of SLCO1B1 allele frequencies the genotype distribution of the SLCO1B1388A>G locus between the two groups(P>0.05),and the genotype distribution at the 521T>C locus was statistically different(P<0.05).Logistic regression analysis showed that SLCO1B1 genotypes were not significantly asso-ciated with the elderly T2DM complicated with MCI(P>0.05),and APOE alleles and APOE genotypes were significantly associated with the elderly T2DM complicated with MCI(P<0.05).Conclusion APOE gene polymorphism is associated with mild cognitive impairment in elderly patients with type 2 diabetes.APOE alleles and genotypes are independent risk factors for elderly type 2 diabetes patients with mild cognitive impairment.SLCO1B1 gene polymorphism is not associated with mild cognitive impairment in elderly patients with type 2 diabetes.
