摘  要:目的探讨芒果苷对亚硒酸钠所致大鼠白内障的改善作用。方法将新生9 d龄的40只Wistar大鼠随机分为4组,分别为对照组(CG)、模型组(MG)、芒果苷低剂量组(L-MAN)、芒果苷高剂量组(H-MAN),每组10只。MG、L-MAN、H-MAN组通过一次性皮下注射2.46 mg/kg亚硝酸钠建立大鼠白内障模型,CG组皮下注射等量0.9%氯化钠注射液。L-MAN、H-MAN组大鼠在造模1 h后,分别进行连续7 d的10、40 mg/kg芒果苷腹腔注射,CG、MG组仅给予等量0.9%氯化钠注射液。21 d后,对大鼠晶状体进行形态学检查,根据晶状体混浊程度进行分级评分,检测大鼠晶状体中钙离子浓度、晶状体总水溶性蛋白、丙二醛(MDA)和还原性谷胱甘肽(GSH)水平。结果(1)晶状体混浊评分:与CG组比较,MG组晶状体混浊评分升高(t=22.136,P=0.000);与MG组比较,L-MAN、H-MAN大鼠晶状体混浊评分降低(t_(L-MAN)=4.833,t_(H-MAN)=7.691,均P=0.000),差异均有统计学意义。(2)晶状体钙离子浓度:与CG组比较,MG组大鼠晶状体钙离子浓度升高(t=22.046,P=0.000);与MG组比较,L-MAN、H-MAN组大鼠晶状体钙离子浓度降低(t_(L-MAN)=13.706,t_(H-MAN)=16.982,均P=0.000),差异均有统计学意义。(3)晶状体总水溶性蛋白:与CG组比较,MG组的大鼠晶状体总水溶性蛋白水平降低(t=8.741,P=0.000);MG组比较,L-MAN、H-MAN组大鼠晶状体总水溶性蛋白水平升高(t_(L-MAN)=3.061,P=0.007;t_(H-MAN)=9.742,P=0.000),差异均有统计学意义。(4)氧化应激:与CG组比较,MG组大鼠晶状体MDA水平升高(t=31.917,P=0.000),GSH水平降低(t=27.540,P=0.000);与MG组比较,L-MAN、H-MAN组大鼠晶状体MDA水平降低(t_(L-MAN)=11.988,t_(H-MAN)=20.427,均P=0.000),GSH升高(t_(L-MAN)=9.229,t_(H-MAN)=33.772,均P=0.000),差异均有统计学意义。结论芒果苷可能通过维持大鼠晶状体内钙离子稳态,抑制亚硒酸钠引起的晶状体内总水溶性蛋白减少,减轻大鼠白内障。OBJECTIVE To investigate the effect of mangiferin on cataract rats induced by sodium selenite.METHODS A total of 409-day-old Wistar rats were randomly divided into four groups:Control group(CG),model group(MG),low-dose mangiferin(L-MAN)group,high-dose mangiferin(H-MAN)group,with 10 rats in each group.In MG,L-MAN and H-MAN groups,the cataract model were established by subcutaneous injection of 2.46 mg/kg sodium nitrite once,while in CG,0.9%sodium chloride injection was injected subcutaneously.One hour after modeling,the rats in L-MAN and H-MAN groups were given 10 and 40 mg/kg mangiferin intraperitoneal injection for seven days consecutively,while the rats in CG and MG were only given 0.9%sodium chloride injection of the same amount.After 21 d,the rat lens was morphologically examined and graded according to the degree of lens opacity.The concentration of calcium ion,total water-soluble protein,malondialdehyde(MDA)and reduced glutathione(GSH)in the rat lens were detected.RESULTS(1)Lens opacity scores:Compared with CG,the lens opacity scores in MG increased(t=22.136,P=0.000);Compared with MG,the lens opacity scores of L-MAN and H-MAN groups decreased(t_(L-MAN)=4.833,t_(H-MAN)=7.691,all P=0.000).The differences were statistically significant.(2)Lens calcium concentration:Compared with CG,the lens calcium concentration of MG increased(t=22.046,P=0.000);Compared with MG,the concentration of calcium ions in lens of L-MAN and H-MAN groups decreased(t_(L-MAN)=13.706,t_(H-MAN)=16.982,both P=0.000).The differences were statistically significant.(3)Total water-soluble protein of lens:Compared with CG,the level of total watersoluble protein of lens in MG decreased(t=8.741,P=0.000);Compared with MG,the level of total water-soluble protein in lens of rats in L-MAN and H-MAN groups increased significantly(t_(L-MAN)=3.061,P=0.007;t_(H-MAN)=9.742,P=0.000).The differences were statistically significant.(4)Oxidative stress:Compared with CG,MDA level in lens of MG increased(t=31.917,P=0.000),GSH level decreased(t=27.540,P=0.000);Com
