摘  要:目的:通过总结CD19蛋白结构与功能相关的研究进展,并结合工程化表达的研究案例,为设计、制备用于CAR-T细胞检测与激活的CD19探针提供策略性指导。方法:结合文献和数据库搜索,从一级序列到高级结构系统介绍了CD19蛋白的结构特征,和其结构改变与临床疾病的关系,总结了CD19工程化改造的关注要点。结果:参考CD19蛋白结构与功能的关系,总结了CD19蛋白工程的不同策略,为(靶向CD19的)CAR-T细胞的检测与激活提供了参考。结论:CD19分子在淋巴瘤B细胞上高表达,是嵌合抗原受体T细胞免疫疗法(Chimeric Antigen Receptor T-Cell Immunotherapy,CAR-T)的重要靶点之一。CD19的表达量和稳定性是其工程化改造的关注要点,可以通过点突变或构建融合蛋白实现。稳定的CD19类蛋白探针是监测CAR-T细胞的有效工具。Aims:This paper summarizes the research progress on the structure and functions of CD19 protein combined with the engineering expression research cases.It will provide strategic guidance for the design and preparation of CD19 probes which are important for the detection and activation of CAR-T cells.Methods:Based on literature and database search,the structural characteristics of CD19 protein were systematically introduced from the primary structure to advanced structure.The relationship between structural changes and clinical disease and the key points of CD19 engineering were summarized.Results:Referring to the relationship between the structure and the function of CD19 protein,different strategies of CD19 protein engineering were concluded,which could provide a reference for the detection and activation of CD19-targeted CAR-T cells.Conclusions:The high expression of CD19 in lymphoma B cells is one of the important targets of chimeric antigen receptor T-cell immunotherapy(CAR-T).The expression level and the stability of CD19 are the main concerns of its engineering,which can be realized by point mutation or constructing fusion proteins.Stable CD19 type protein probes are effective for detecting CAR-T cells.
