摘 要:本研究对青海省内采集的牦牛和藏羊粪便样本中线虫进行了锚定高通量测序检测和PCR检测验证。结果表明,锚定高通量测序结果显示有效物种OTU共156条。物种数据库注释的有33种生物,包括已知动植物线虫(37.34%)、已知细菌(10.44%)和未知生物种(2.61%)。其中已知的动物线虫以钩口属为主,其次是血矛属。针对钩虫,PCR检测结果显示:海晏县阳性率最高(25%),其次是泽库县(8.82%)和河南县(6.9%),贵南县样本中钩虫阳性率为0。结果提示本研究中的锚定高通量测序方法的检测结果中尽管存在细菌,但客观上起到了缩小被检物种范围的作用,这为开展群体性高通量寄生虫检测的深入研究提供了参考资料。In this study, the nematodes in fecal samples of yak and Tibetan sheep were detected by anchored high-throughput sequencing and PCR detection and verified. The results showed that there were total of 156 effective species OTUs by anchored high-throughput sequencing, and there were 33 species of organisms annotated in the species database, including plant and animal nematodes(37.34%),bacteria(10.44%) and unknown species(2.61%). Among the known animal nematodes, Ancylostoma was the main species, followed by Haemonchus. For hookworm, the PCR detection results showed that the highest positive rate(25%) occur in Haiyan County,followed by Zeku County(8.82%) and Henan County(6.9%),and the positive rate of hookworm in samples from Guinan County was 0. The results suggest that although there were bacteria in the detection results of the anchored high-throughput sequencing method in this study, which objectively played a role in narrowing the range of the detected species, which provided a reference for in-depth research on the high-throughput parasite detection.
