摘 要:目的优化蒲地灌肠液的提取工艺。方法以芍药苷、咖啡酸的含量和干浸膏得率为评价指标,采用正交设计试验,考察浸泡时间、料液比和提取总时间对提取结果的影响,确定蒲地灌肠液的最佳提取工艺。结果蒲地灌肠液的最佳工艺条件为浸泡时间20 min,提取2次,每次14倍量溶剂,提取总时间为90 min,该提取工艺下得到的芍药苷含量、咖啡酸含量和干浸膏得率均值分别为663.4μg·ml^(-1)、46.02μg·ml^(-1)和20.34%。结论多指标综合评分法联合正交设计优化蒲地灌肠液提取工艺符合中药多成分多靶点的作用特点,优化的提取工艺稳定可行。Objective To optimize the extraction process of Pudi enema.Methods The contents of paeoniflorin,caffeic acid and the yield of dry extract were used as evaluation indexes.Orthogonal design was used to determine the effects of such factors as the duration of soaking,solid-liquid ratio and total duration of extraction on the extraction results and to select the optimal extraction process of Pudi enema.Results The optimum extraction process of Pudi enema was a soaking time of 20 minutes,two-time extraction,14 times as much dilution each time,and the total duration extraction of 90 minutes.The contents of paeoniflorin and caffeic acid and yield of dry extract were 663.4μg·ml^(-1),46.02μg·ml^(-1)and 20.34%,respectively.Conclusion The optimization of the extraction process with the multi-index comprehensive scoring method is compatible with the multi-component and multi-target characteristics of traditional Chinese medicines,and the optimized extraction process is stable and feasible.
