摘 要:目的 观察参附注射液对心肌缺血再灌注大鼠血清炎性因子及Toll样受体4(TLR4)、核因子(NF)-κB的影响,探讨可能的作用机制。方法 将60只雄性SD大鼠随机分为对照组(10只)、假手术组(10只)、模型组(20只)、参附组(20只)。分别检测各组大鼠血清肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)、C反应蛋白(CRP)水平及心肌组织TLR4、NF-κB mRNA及蛋白表达水平。结果 模型组血清TNF-α、IL-6、CRP表达水平较对照组升高(P<0.05);参附组TNF-α、IL-6、CRP表达水平较模型组下降(P<0.05)。模型组NF-κB、TLR4蛋白表达较对照组升高(P<0.05);参附组NF-κB、TLR4蛋白表达较模型组下降(P<0.05)。模型组NF-κB、TLR4 mRNA表达较对照组升高(P<0.05);参附组NF-κB、TLR4 mRNA表达水平较模型组下降(P<0.05)。结论 参附注射液对心肌缺血再灌注损伤大鼠有保护作用,其机制可能与调控TLR4/NF-κB通路、抑制炎症反应有关。Objective To observe the effects of Shenfu Injection on serum inflammatory factors, Toll-like receptor 4(TLR4),and nuclear factor(NF)-κB in myocardial ischemia-reperfusion rats.Methods Sixty male SD rats were randomly divided into the control group(10 rats),sham group(10 rats),model group(20 rats),and Shenfu group(20 rats).Serum tumor necrosis factor α(TNF-α),interleukin-6(IL-6),C-reactive protein(CRP) and myocardial tissue TLR4,NF-κB mRNA and protein expression of rats were detected respectively.Results The levels of serum TNF-α,IL-6 and CRP in the model group were significantly increased than those in the control group(P<0.05),while the levels of TNF-α,IL-6 and CRP in the Shenfu group were significantly decreased than those in the model group(P<0.05).The protein expressions of NF-κB and TLR4 in the model group were increased than those in the control group(P<0.05),while the protein expressions of NF-κB and TLR4 in the Shenfu group were decreased than those in the model group(P<0.05).The mRNA expressions of NF-κB and TLR4 in the model group were increased than those in the control group(P<0.05),while the mRNA expressions of NF-κB and TLR4 in the Shenfu group were decreased than those in the model group(P<0.05).Conclusion Shenfu Injection showed protective effect on myocardial ischemia-reperfusion injury in rats, and its mechanism might be related to regulating TLR4/NF-κB pathway and inhibiting inflammation response.
