常乐康对早产小鼠CD8+T细胞CD28和PD-1表达的实验研究

摘  要:目的:探讨常乐康早期干预对早产小鼠外周血CD8+T细胞表达CD28和PD-1的影响。方法:随机选取孕18 d的孕鼠,用米福RU486诱导早产,获得胎龄<20 d的早产小鼠,随机分为早产实验组(A组)和早产对照组(B组);随机选择部分孕鼠自然分娩,获得足月新生小鼠为足月对照组(C组),每组20只。A组自生后第1天起予以常乐康进行灌胃,B、C组用等量生理盐水灌胃,连续1周。在生后第14天和第21天,取外周血用流式细胞仪检测CD8+T细胞表面CD28和PD-1的百分率及其平均荧光强度(MFI)。结果:①第14天时,A组CD28百分率高于C组(P<0.05),但低于B组(P<0.05);第21天时,C组高于A、B组(P<0.05)。第14天时,A组CD28 MFI高于B、C组(P<0.05);第21天时,A组高于B组(P<0.05),但低于C组(P<0.05)。②第14天时,A组PD-1百分率均高于B、C组(P<0.05);第21天时,B组高于A、C组(P<0.05)。第14天时,A组PD-1 MFI高于B、C组(P<0.05);第21天时,A、C组高于B组(P<0.05)。结论:常乐康的干预能促进早产小鼠免疫系统的发育和成熟,其生物学作用可能是通过对CD8+T细胞表面CD28和PD-1表达的调控。Objective:To investigate the effect of early intervention using Changlekang live probiotic powder on the expression of CD28 and PD-1 in peripheral blood CD8+T cells in preterm mice.Methods:The 18-day gestational mice were randomly selected,and were induced for preterm birth using RU486.Preterm mice with gestational age<20 days were obtained,and then randomized to group A(preterm birth for experimental observation)and group B(control group).Next,pregnant mice were randomly selected to undergo spontaneous labor.The newborn mice with gestational age>20 days were included in group C(full-term control,n-20 for each group).Mice in group A received administration using Changlekang live probiotic powder,and those in group B and C were treated with the same amount of normal saline by gavage for one week at day one after birth.By the 14 th and the 21 st day after birth,peripheral blood was collected in all mice to detect the percentage of CD28 and PD-1 on the surface of CD8+T cells and the fluorescence intensity(MFI)of the two co-stimulatory molecules by flow cytometry.Results:①By the 14 th day,the percentage of CD28 was higher in group A than in group C(P<0.05),yet lower than in group B(P<0.05).By the 21 st day,the percentage of CD28 was higher in group C than in group A and group B(P<0.05).Group A had higher CD28 MFI than group B and group C at day 14(P<0.05).CD28 MFI was higher in group A than in group B(P<0.05),yet lower than in group C at day 21(P<0.05);②By the 14 th day,the percentage of PD-1 was higher in group A than in group B and group C(P<0.05).Group B had higher PD-1 level than group A and group C by the 21 st day(P<0.05).At day 14,PD-1 MFI was higher in group A than in group B and C(P<0.05),yet was higher in group A and group C than in group B at day 21(P<0.05).Conclusion:Early supplementation with Changlekang live probiotic powder may contribute to development and maturation of immune system in preterm mice by regulating the expression of CD28 and PD-1 on CD8+T cells.
