摘 要:目的 观察电针对乳腺增生(MGH)大鼠乳房形态(乳腺组织、乳头直径、高度)和血清促性腺激素释放激素(GnRH)、促卵泡刺激素(FSH)、促黄体生成素(LH)、泌乳素(PRL)这4种性激素的影响,从下丘脑-垂体水平探讨电针治疗MGH的机制。方法 选雌性健康未孕SD大鼠50只,在适应性喂养后随机分成空白组和造模组,其中造模组造模成功后分模型组、溴隐亭组、电针组。溴隐亭组应用溶在氯化钠的溴隐亭片溶液0.393 mg/kg灌胃治疗。电针组选郭诚杰教授经验用穴(甲组、乙组)交替用电针治疗。分组治疗均为1次/d, 1个疗程5次,总共4个疗程,每疗程间隔2 d。结束后对大鼠第二对乳头高度、直径测量,取第二对乳房乳腺组织并进行病理学检察;腹主动脉采血、离心、取血清,ELISA检测下丘脑GnRH和垂体FSH、LH、PRL含量。结果 模型组大鼠乳房形态符合增生表现,血清LH降低,GnRH、FSH、PRL水平升高,和空白组相比差异有统计学意义;电针组、溴隐亭组、模型组相比差异有统计学意义;溴隐亭组和电针组相比差异无统计学意义。结论 电针可明显改善MGH大鼠增生的乳腺组织,降低乳头高度、减小乳头直径;降低MGH大鼠血清GnRH、FSH、PRL含量,升高LH含量,和溴隐亭治疗效应相似。Objective To observe the effects of electroacupuncture on breast morphology(breast tissue, nipple diameter, height) and serum gonadotropin-releasing hormone(GnRH),follicle stimulating hormone(FSH),luteinizing hormone(LH),prolactin(PRL) in rats with mammary hyperplasia(MGH) and explore the mechanism of electroacupuncture treating MGH from the hypothalamus pituitary level.Methods Fifty healthy female non-pregnant SD rats were selected and randomly divided into blank group and model group after adaptive feeding.The model group was divided into model group, bromoyntine group and electroacupuncture group after successful model building.Bromophentine group was treated with 0.393 mg/kg solution of bromophentine tablets dissolved in sodium chloride by gavage.In the electroacupuncture group, professor GUO Chengjie′s experience points(group A,group B) were treated with electroacupuncture alternately.The treatment was given 1 time/day.There was 1 course of 5 times, a total of 4 courses, and each course interval was 2 days.At the end, the height and diameter of the second pair of nipples were measured, and the mammary tissue of the second pair of breasts was taken out for the pathological examination.Blood samples were collected from abdominal aorta, centrifuged and serum samples were collected to detect GnRH content in hypothalamus and the levels of FSH,LH and PRL in pituitary by ELISA.Results Breast morphology of the model group was consistent with hyperplasia, serum LH decreased, the levels of GnRH,FSH and PRL increased, and there were significant differences compared with those of the blank group.There were significant differences among electroacupuncture group, bromocriptine group and model group.There was no significant difference between the bromocriptine group and electroacupuncture group.Conclusion Electroacupuncture can obviously improve the hyperplasia mammary tissue, reduce the height and diameter of nipples in MGH rats.The contents of GnRH,FSH and PRL in serum of MGH rats were decreased, while the contents of
