摘 要:目的 探讨痛风肾泰方逆转非结晶尿酸导致的人肾小管上皮细胞转分化的作用机理。方法 采用体外非结晶尿酸培养HK-2细胞造模,痛风肾泰方大鼠灌胃制备含药血清,采用放射免疫法检测培养液内生长转化因子-β1(TGF-β1)浓度,RT-PCR检测HK-2细胞TGFβRImRNA、TGFβRIImRNA、α平滑肌肌动蛋白(α-SMA)mRNA、E-cadherin mRNA、Smad3mRNA、Smad7mRNA含量。结果 与对照组比较,尿酸组TGF-β1浓度、TGFβRImRNA、α-SMA mRNA、Smad3 mRNA升高、E-cadherin mRNA降低,具有统计学意义,TGFβRIImRNA、Smad7轻度升高,结果无统计学意义;与尿酸组比较,中药组TGF-β1浓度、α-SMA mRNA、Smad3 mRNA降低,具有统计学意义,E-cadherin升高、TGFβRI、TGFβRIImRNA降低,无统计学意义,Smad7具有明显的升高作用,与对照组比较差异有统计学意义。结论 非结晶尿酸可以通过TGF-β1-TGFβR-Smad途径导致HK-2细胞EMT,中药痛风肾泰方含药血清具有抑制该通路、逆转HK-2细胞EMT发生的作用。Objective To investigate the mechanism of Tongfeng Shentai Recipe(痛风肾泰方)reversing human renal tubule epithelial cells epithelial-mesenchymal transition(EMT)caused by non-crystallized uric acid.Methods HK-2 cells were cultured with non-crystallized uric acid in vitro,and the drug-containing serum was prepared by intragastric administration in rats with Tongfeng Shentai Recipe.The contents of transforming growth factor-β1(TGF-β1)in the medium was detected by radioimmunoassay.The mRNA contents of TGFβRImRNA,TGFβRIImRNA,αsmooth muscle actin(α-SMA)mRNA,E-cadherin mRNA,Smad3 mRNA and Smad7 mRNA in HK-2 cells were detected by RT-PCR.Results Compared with those of the control group,the contents of TGF-β1,TGFβRImRNA,α-SMA mRNA,Smad3 mRNA in uric acid group were increased and the content of E-cadherin was decreased,with statistical significance.TGFβRIImRNA and Smad7 were slightly increased,and the results were not statistically significant.Compared with those of the uric acid group,the contents of TGF-β1,α-SMA mRNA and Smad3 mRNA in Tongfeng Shentai Recipe group were decreased with statistical significance.The content of E-cadherin was increased and the contents of TGFβRI and TGFβRIImRNA were decreased without statistical significance.Compared with those of the control group,Smad7 had a significant elevation.Conclusion Non-crystalline uric acid can induce EMT in HK-2 cells through TGF-β1-TGFβR-Smad pathway,and the serum containing Tongfeng Shentai Recipe can inhibit this pathway and reverse EMT of HK-2 cells.
