摘 要:目的研究人参根饮液对免疫功能低下模型小鼠的免疫调节作用及潜在的作用机制。方法将SPF级KM小鼠随机分为空白组、模型组、人参根饮液高剂量组(10 mL/kg)、人参根饮液低剂量组(5 mL/kg)、预防组(10 mL/kg)。除空白组外,各组小鼠采用皮下注射联合肌肉注射氢化可的松的方法进行免疫功能低下复制。以免疫器官脏器指数、巨噬细胞吞噬指数、脾淋巴细胞增殖指数、白细胞和中性粒细胞含量、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)含量等为评价指标,评价人参根饮液对免疫功能低下模型小鼠的作用。在数据库中检索人参主要化学成分作用靶点和免疫低下相关靶点,用韦恩图得到交集靶点。运用Cytoscape 3.7.1软件构建交集靶点的蛋白相互作用(protein-protein interaction,PPI)网络图和“中药-成分-靶点”网络图,筛选出度值高的关键靶点和成分。将交集靶点上传Metascape网站进行基因本体(gene ontology,GO)功能富集分析和京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)富集分析,预期其作用机制。结果与模型组相比,人参根饮液高剂量组、预防组明显的回调小鼠脏器指数(P<0.01),提高小鼠血清中免疫因子的值(P<0.01),增强小鼠巨噬细胞吞噬能力(P<0.05),增强小鼠脾淋巴细胞的增殖能力(P<0.05),提升小鼠白细胞、中性粒细胞的含量(P<0.01)。人参中筛选出22个有效成分,110个靶点,免疫低下疾病靶点2195个,药物与疾病相关靶点108个。筛选得出人参根饮液中山柰酚、β-谷甾醇、豆甾醇等成为核心效应成分,可作用于PTGS2、PTGS1、ADRB2等重要靶点,靶点涉及到的关键通路有糖尿病并发症中的AGE-RAGE信号通路、TNF信号通路、IL^(-1)7信号通路等。结论人参根饮液可显著提升模型小鼠免疫功能,其作用机制具有多靶点、多通路的特点。Objective To clarify the effect of Ginseng Root Drink on immune regulation and its potential mechanism.Methods SPF grade KM mice were randomly divided into blank group,model group,high-dose Ginseng Root Drink group(10 mL/kg),low-dose group(5 mL/kg)and prevention group(10 mL/kg).Except for the blank group,mice in each group were replicated by subcutaneous injection combined with intramuscular injection of hydrocortisone.The immune organ index,macrophage phagocytosis index,spleen lymphocyte proliferation index,leukocyte and neutrophil content,IL-6 and TNF-αcontent were used as evaluation indexes to evaluate decreasing the immune function of Ginseng Root Drink to model mice.The target of the main chemical components of ginseng and the targets related to immunocompromised were searched in the database,and the intersection targets were obtained by Venn diagram.Cytoscape 3.7.1software was used to construct the protein-protein interaction(PPI)network diagram of the intersection targets and the“traditional Chinese medicine-component-target”network diagram,and screen out the key targets and components with high degree value.The intersection targets were uploaded to the Metascape website for gene ontology(GO)functional enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG)enrichment analysis to predict their mechanism of action.Results Compared with the model group,the high-dose Ginseng Root Drink group and the prevention group significantly adjusted the organ index of the mice(P<0.01),increased the value of immune factors in the serum of the mice(P<0.01),and enhanced the macrophages of the mice.The phagocytic ability of phagocytes(P<0.05),the proliferation ability of spleen lymphocytes in mice(P<0.05),and the content of leukocytes and neutrophils in mice(P<0.01).From ginseng,22 active ingredients and 110 targets were screened,2195 targets for immunocompromised diseases,and 108 targets for drugs and diseases.It was screened that kaempferol,β-sitosterol,stigmasterol,etc.in Ginseng Root Drink became the
