摘 要:目的探究雌激素及其抑制剂氟维司群对子宫内膜异位症(EMs)在位子宫内膜间质细胞神经生长因子(NGF)、丝裂原活化蛋白激酶(MAPK)及环腺苷酸应答元件结合蛋白(CREB)表达的影响。方法收集EMs患者9例,分离培养子宫内膜间质细胞,应用免疫荧光染色检测子宫内膜间质细胞中NGF、MAPK及CREB的表达及定位情况;采用Western blot及qRT-PCR法检测雌激素及其抑制剂氟维司群干预后子宫内膜间质细胞中NGF、MAPK及CREB蛋白及mRNA表达变化情况。结果细胞免疫荧光染色结果显示子宫内膜间质细胞中NGF、MAPK及CREB蛋白均主要定位于胞浆中;Western blot结果显示,与对照组相比,EMs患者在位内膜间质细胞中NGF、MAPK及CREB的表达升高(P均<0.05)。Western blot及qRT-PCR检测结果显示,与对照组相比,雌激素作用后,子宫内膜间质细胞中NGF、MAPK及CREB蛋白及mRNA表达水平均上升,氟维司群作用后NGF、MAPK及CREB蛋白及mRNA表达水平均下降(P均<0.01)。结论EMs中NGF、MAPK及CREB蛋白表达升高,揭示其可能与EMs相关疼痛发生密切相关,并受雌激素药物调节,可作为EMs疼痛治疗中的一个靶点。Objective To investigate the effects of estrogen and its inhibitors on the expression of nerve growth factor(NGF),mitogen-activated protein kinase(MAPK)and cyclic adenylate response element binding protein(CREB)in interstitial cells of patients with EMs.Methods Nine cases of EMs eutopic endometrium were collected,EMs eutopic endometrium stromal cells were isolated and cultured.Immunofluorescence staining was used to detect the localization of NGF,MAPK and CREB in the mesenchymal cells of patients with EMs.Western blot and qRT-PCR were used to detect the changes in the expression of NGF,MAPK and CREB in interstitial cells of EMs patients after the intervention of estrogen and its inhibitors.Results The results of cellular immunofluorescence staining showed that NGF,MAPK and CREB proteins in endometrial stromal cells were mainly located in the cytoplasm;Western blot showed that compared with the control group,the expression of NGF,MAPK and CREB in eutopic endometrial stromal cells of EMs patients increased(P all<0.05).However,Western blot and qRT-PCR results showed that the expression of NGF,MAPK and CREB in eutopic endometrial stromal cells of EMs patients increased after estrogen treatment,while decreased after fluvison treatment(P<0.01).Conclusion The occurrence of endometriosis pain may be closely related to the expression of NGF,MAPK and CREB,and is regulated by estrogen,which may serve as a target in the treatment of endometriotic pain syndrome.
