摘 要:为了研究不同方法提取的桔梗多糖对小鼠体内抗氧化性能的影响,试验采用水提法和微波法提取桔梗多糖,分析桔梗多糖的理化性质;分别用两种方法提取的不同剂量的桔梗多糖灌胃小鼠,并设空白对照组及阳性对照组,连续灌胃16 d,分别在灌胃前、灌胃第8天、处死前对各组小鼠称重,分析桔梗多糖对小鼠体重的影响,试验结束后处死小鼠,计算肝脏和脾脏指数,测定灌胃第16天桔梗多糖的小鼠肝脏、血浆中丙二醛(MDA)含量和超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性。结果表明:两种方法提取的桔梗多糖均为乳白色固体粉末,组织结构致密,理化性质无明显差别。与空白对照组相比,两种提取方法的高剂量组桔梗多糖均能极显著增加小鼠的平均日增重、肝脏指数和脾脏指数(P<0.01);水提多糖高剂量组和微波多糖高、中剂量组的血浆、肝脏中SOD活性均极显著高于空白对照组(P<0.01);微波多糖各剂量组血浆中的GSH-Px活性均极显著高于空白对照组(P<0.01),水提多糖高、中剂量组和微波多糖高剂量组肝脏中的GSH-Px活性极显著高于空白对照组(P<0.01)。水提多糖高剂量组和微波多糖高、中剂量组的小鼠血浆、肝脏中MDA含量均极显著低于空白对照组(P<0.01)。说明两种方法提取的桔梗多糖外观形态和理化性质无明显差别,均能提高小鼠平均日增重、肝脏指数、脾脏指数和抗氧化能力,其中微波高剂量组的抗氧化效果最佳。In order to study the effects of different methods of extracting Platycodon grandiflorus polysaccharides on antioxidant properties in mice, aqueous and microwave methods were used to extract Platycodon grandiflorus polysaccharides and to analyze the physicochemical properties of Platycodon grandiflorus polysaccharides. Mice were gavaged with different doses of Platycodon grandiflorus polysaccharides extracted by two methods respectively, and a blank control group and a positive control group were set up. The mice were gavaged continuously for 16 days. The mice in each group were weighed before gavage, 8 days after gavage and before execution to analyze the effect of Platycodon grandiflorus polysaccharides on their body weights. At the end of the experiment, mice were executed;liver and spleen indices were calculated, and malondialdehyde(MDA) content, superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) activities in liver and plasma of mice gavaged with Platycodon grandiflorus polysaccharides for 16 days were measured. The results showed that the Platycodon grandiflorus polysaccharides extracted by both methods were creamy white solid powder with dense organization structure with no significant difference in physicochemical properties. Compared with the blank control group, the mean daily weight gain, liver index and spleen index of mice were highly significantly increased(P<0.01) in the high-dose groups of both extraction methods of Platycodon grandiflorus polysaccharides. The SOD activities in plasma and liver of the high dose group of water-extracted polysaccharides and the high and medium dose groups of microwave polysaccharides were highly significantly higher than those of the blank control group(P<0.01). The GSH-Px activity in the plasma of all dose groups of microwave polysaccharides was extremely significantly higher than that of the blank control group(P<0.01);the GSH-Px activity in the liver of the high and medium dose groups of water-extracted polysaccharides and the high dose group of microwa
